| Objective As a new type of muscle cytokine,Irisin is produced by cutting the membrane protein FNDC5.FNDC5/Irisin is involved in the regulation of fat browning,metabolism,immunity and chronic inflammation.FNDC5/Irisin is closely related to a variety of common malignant tumors.However,its function and underlying mechanism in the development of liver cancer are still unclear.In this study,we aim to study the expression of FNDC5/Irsin in hepatocellular carcinoma and its effect on liver cancer cells,so as to provide experimental evidence for the further clinical application of FNDC5/Irisin.Method1.Check the expression of FNDC5/Irisin in normal liver cells and hepatocellular carcinoma cells by Western blotting,and use immunohistochemistry to evaluate the expression of FNDC5/Irisin in 52 cases of hepatocellular carcinoma patients and corresponding adjacent tissue samples,and observe the expression of FNDC5/Irisin The relationship between expression and clinical characteristics.2.Use Cell Counting Kit 8(CCK-8)to examine the influence of Irisin on the cell proliferation of SMCC7721 and Hep G2 hepatocellular carcinoma cells,and detect cell migration and invasion by cell scratch test and Transwell invasion test.3.In addition,Irisin was used to examine the expression of epithelial mesenchymal transition(EMT)markers in hepatocellular carcinoma cells and its effect on AKT and pAKT proteins in the PI3K/AKT pathway by Western blotting assay.Results1.The expression level of FNDC5/Irisin protein in hepatocellular carcinoma cells cells and hepatocellular carcinoma tissues is significantly higher than that of normal cells and adjacent normal tissues.The expression of FNDC5/Irisin is related to hepatocellular carcinoma microvascular metastasis.2.CCK-8 experiments show that Irisin promotes the proliferation of hepatocellular carcinoma cells.Scratch test and Transwell test show that Irisin promotes the migration and invasion of hepatocellular carcinoma cells3.Western blot experiments show that Irisin can promote the expression of EMT markers in hepatocellular carcinoma cells,and promote the expression of p-AKT protein in hepatocellular carcinoma cells,while the total protein AKT level has no significant change.Conclusions1.The expression level of FNDC5/Irisin protein in hepatocellular carcinoma cells and hepatocellular carcinoma tissues is significantly higher than that of normal liver cells and adjacent tissues.The expression of FNDC5/Irisin is related to hepatocellular carcinoma microvascular metastasis.2.Irisin can promote the cell proliferation of hepatocellular carcinoma cells,and at the same time promote the migration and invasion of hepatocellular carcinoma cells.3.Irisin can promote the occurrence of EMT of hepatocellular carcinoma cells by promoting the activation of the PI3K/AKT/Snail pathway of hepatocellular carcinoma cells. |
