| Objectives:To observe the effect of 6-gingerol on gastric residual rate and intestinal propulsive rate in chemotherapy rats,and investigate the mechanisms of 6-gingerol alleviating gastrointestinal motility disorder in chemotherapy rats from the perspectives of gastrointestinal hormones and interstitial cells of Cajal(ICC).Methods:The male SD rats were randomly divided into seven groups: normal control group,6-gingerol normal control group,model group,positive drug group,and 6-gingerol low,medium and high-dose groups.The rats of 6-gingerol control group and the 6-gingerol low,medium and high-dose groups were administered with 6-gingerol with 100,25,50 and100 mg/kg,respectively.The rats of positive drug group were administered with metoclopramide with 3 mg/kg.The rats of normal control group and model group were administered with an equal volume of solvent.The above dose was administered once a day for 4 consecutive days.One hour after administered on the first day,the rats of the model group,the positive drug group,and the 6-gingerol low,medium,and high-dose groups were intraperitoneally injected with cisplatin with 6mg/kg.The rats of the normal control group and the 6-gingerol normal control group were injected with an equal volume of normal saline.After48 hours of modeling,rats were deprived of food for 24 hours.All rats were administered with the nutritional paste after 72 hours of modeling,and the stomach and small intestine were taken to detect the stomach contents,gastric residual rate and small intestinal propulsive rate after 30 minutes.The levels of motilin(MTL),gastrin(GAS),vasoactive intestinal peptide(VIP),somatostatin(SS)in serum were detected by ELISA.Tissue from antrum and ileum were stained with hematoxylin eosin(HE)to observe pathological changes.The m RNA expression levels of TNF-α,IL-6,c-kit,SCF in gastric antrum and ileum were detected by real-time PCR.The protein levels of c-kit and SCF in gastric antrum and ileum were detected using Western blot,the distribution of c-kit in gastric antrum and ileum was detected by immunohistochemistry.Results:1 、 Effect of 6-gingerol on weight of gastric contents and gastric residual rateThe weight of gastric contents and gastric residual rate in the model group were significantly increased compared with the normal control group(P < 0.01).Compared with the model group,the weight of gastric contents and gastric residual rate in the 6-gingerol high-dose group were significantly decreased(P < 0.01,P < 0.05).2、Effect of 6-gingerol on small intestinal propulsive rateThe small intestinal propulsive rate of rats administered with cisplatin was significantly increased compared with the normal control group(P < 0.05).Compared with the model group,the small intestinal were observed in the model group,and the pathological changes in gastric antrum and ileum of the 6-gingerol high-dose group were alleviated.5、Effects of 6-gingerol on m RNA expression levels of TNF-α,IL-6,c-kit and SCF in gastrointestinal tractCisplatin administration significantly increased m RNA expression levels of TNF-α and IL-6(P < 0.01),and the m RNA expression levels of c-kit and SCF in gastric antrum and ileum were significantly decreased(P< 0.01),compared to the normal control group.Compared with the model group,6-gingerol significantly decreased the m RNA expression levels of TNF-α and IL-6,increased m RNA expression levels of c-kit and SCF in gastric antrum and ileum(P < 0.01).6、Effect of 6-gingerol on protein expression levels of c-kit and SCF in gastrointestinal tractCompared with the normal control group,the protein expression of c-kit and SCF proteins in gastric antrum and ileum of the model group were significantly decreased(P < 0.01).6-gingerol significantly increased protein expression levels of c-kit and SCF in gastric antrum and ileum,compared with the model group(P < 0.01).7 、 Effect of 6-gingerol on c-kit immunohistochemistry of gastrointestinal tissueAfter cisplatin administration,the average optical density of c-kit in gastric antrum and ileum was significantly decreased(P < 0.01).Compared with the model group,the average optical density of c-kit in gastric antrum and ileum in the 6-gingerol high-dose group was significantly increased(P < 0.01).Conclusion:6-gingerol can ameliorate gastrointestinal motility disorders in chemotherapy rats,which may be achieved by protecting gastrointestinal ICC and regulating the levels of gastrointestinal hormones through the effects of anti-inflammation. |
