| Fallopia denticuta is the root of Polygonum multiflorum plant [Fallopia denticuta(Huang)A.J.Li].It is a commonly used Chinese herbal medicine of Yunnan Yi nationality.Fallopia denticuta has a good effect on dysentery,diarrhea,dyspepsia,chronic hepatitis,gastric diseases and other diseases,especially in the treatment of gastric diseases.However,there are currently few studies on its mechanism and pharmacological active ingredients in the treatment of gastric diseases.Therefore,This article studies the anti-gastric ulcer activity,antibacterial activity and chemical components of Fallopia denticuta,which provides a theoretical basis for exploring the pharmacological activity of Fallopia denticuta and its relationship with chemical components.Details as follows:1.The protective effect of Fallopia denticuta extract on human gastric epithelial cells(GES-1).1% ethanol was used to model GES-1 cells,and the success of the model was judged by the degree of cell damage and death.After the model was successfully built,the repair effects of different Fallopia denticuta extracts on GES-1 cells were observed.The results found that different Fallopia denticuta extracts can significantly increase the cell proliferation rate of ethanol-induced GES-1cells(P<0.05),and reduce the damage effect of ethanol on cells.Among them,the 95% ethanol extract of Fallopia denticuta has the best protective effect on GES-1 cells.2.The inhibitory effect of Fallopia denticuta extract on gastric ulcer in rats.The SD rats were divided into a negative control group,a model group,a ranitidine group,and a low,medium and high dose of Fallopia denticuta extract group.A model of rat gastric ulcer induced by ethanol was established.After successful modeling,the morphology of the gastric mucosa and the ulcer condition of the rat were compared,and the ulcer area and ulcer inhibition rate were calculated.The contents of inflammatory factors TNF-α,IL-1β and IL-6 in serum and the contents of antioxidant enzymes SOD,CAT,GSH and MDA in gastric tissue were measured.Western blotting was used to detect the changes of p-p65,p65,p-IκBα,IκBα,p-p38,p38,p-JNK,JNK protein in rat gastric tissue,to explore the activity and mechanism of Fallopia denticuta’s anti-gastric ulcer.There were large areas of injury and ulcers on the gastric mucosal surface of rats in the model group,indicating that the model was successful,while the area of ulcers in each concentration group of the Fallopia denticuta and the positive drug group was significantly reduced(P<0.05).The contents of TNF-α,VIII IL-1β and IL-6 in serum and MDA in gastric mucosal tissues in each concentration group of the Fallopia denticuta and positive drug group were significantly lower than those of the model group,and the contents of SOD,CAT,and GSH in gastric mucosal tissue were significantly higher than those in the model group(P<0.05).Compared with the negative control group,the ratios of p-p65/p65,p-IκBα/IκBα,p-p38/p38,and p-JNK/JNK protein in the gastric tissue of the model group were significantly increased(P<0.05),while in each concentration of Fallopia denticuta group and the positive drug group those were significantly reduced(P<0.05).According to the first part of the three kinds of Fallopia denticuta extract activity screening,namely,the results of the GES-1 cell experiment,the Fallopia denticuta 95% ethanol extract has the best effect,so this section uses the Fallopia denticuta 95% ethanol extract as the test substance to study the effect and mechanism of Fallopia denticuta in inhibiting gastric ulcer in rats.3.Determination of the activity of Fallopia denticuta extract in inhibiting Helicobacter pylori,the pathogenic bacteria of gastric ulcer,and the antibacterial activity of other three common pathogenic bacteria.The antibacterial activity of the three Fallopia denticuta extracts and their different extracts against the four pathogenic bacteria was determined by the punch method,and their MIC and MBC were determined by the double test tube dilution method.It was found that the50% alcohol extract,95% alcohol extract,and decoction extract of Fallopia denticuta have the effect of inhibiting four common pathogenic bacteria,and the 50% alcohol extract of Fallopia denticuta ethyl acetate has the strongest effect,which has the best activity of inhibiting Helicobacter pylori,with MIC of 0.22 mg/m L and MBC of 0.43 mg/m L.4.Determination of the five contents of ingredients in the Fallopia denticuta.The content of gallic acid,catechin(+),cannabiamide F,ethyl protocatechuate,and Throne-1-C-eugenol glycerol in the Fallopia denticuta was determined by HPLC.Chromatographic conditions: 0 ~ 15 min,10 ~30% methanol;15 ~ 35 min,30 ~ 40% methanol;35 ~ 50 min,40 ~ 70% methanol;50 ~ 55 min,70~ 100% methanol.This HPLC method has good linearity,accuracy,precision,and repeatability.The total content of the five components in the three Fallopia denticuta extracts is 0.81,0.74,and 0.74%.5.LC-MS qualitatively and quantitatively detect the compounds in the Fallopia denticuta.Chromatographic conditions: 0 ~ 11.0 min,5 ~ 95%,11.0 ~ 12.0 min,95%.LC-MS detected that there are 236 kinds of compounds in the extract of Fallopia denticuta,mainly flavonoids(35.5%),alkaloids(26.2%),phenolic acids(17.6%),tannins(10.0%),and the content The sum reached89.3%.Using secondary metabolomics analysis,it was found that there are differences in the composition,content and mechanism of action of the secondary metabolites in the three kinds of Fallopia denticuta extracts. |
