Objective:To test the expression level of CUL3(Cullin3)m RNA and protein in peripheral blood mononuclear cells of Kazak in Xinjiang with hypertension,to analyze and explore the relationship between the expression of CUL3 and the pathogenesis of essential hypertension in Kazakh nationality.Methods:An epidemiological investigation was carried out in Hankazitan Township,Manas County,Xinjiang from September 2018 to November 2018,create hypertension database.The case group was randomly selected 24 Kazakh patients with essential hypertension and the control group was 24 health workers of the same nationality.Measure weight,blood pressure,height,triglyceride,high density lipoprotein,low density lipoprotein,total cholesterol,fasting blood glucose,uric acid,creatinine and so on.Extraction of peripheral blood mononuclear cells by Ficoll density gradient centrifugation,Real-time quantitative PCR was used to detect the m RNA level of CUL3 in peripheral blood mononuclear cells,At the same time,the expression of CUL3 protein in mononuclear cells was detected by Westernblot method.To compare the difference of m RNA expression and protein expression of CUL3 measured between the two groups,to analyze the correlation between CUL3 expression level and clinical factor.Results:Differences in age,gender,smoking,triglyceride,fasting blood glucose,total cholesterol,high-density lipoprotein cholesterol were not statistically significant between essential hypertension group and control group.The levels of LDL,BMI,Scr and UA in hypertension group were significantly higher than those in control group(all P < 0.05).The expression of CUL3 m RNA in mononuclear cells of the hypertension group was lower than that of the control group(P < 0.05),there are statistical differences.There was no significant difference in the expression of CUL3 protein in mononuclear cells between the case group and the control group(P > 0.05).Correlation matrix analysis showed that the expression of CUL3 m RNA was negatively correlated with SBP(P < 0.05),BMI was positively correlated with SBP,DBP,UA,TC and LDL-C(all P < 0.05),DBP was positively correlated with LDL-C and UA(all P < 0.05),UA was positively correlated with BMI,DBP,TC and TG(all P < 0.05).Through the analysis of logistic regression analysis,it was found that the expression of CUL3 m RNA,BMI and the level of UA were the risk factors of Kazakh patients with essential hypertension.Conclusion:The decreased expression of CUL3 m RNA in peripheral blood may be related to essential hypertension in Kazakh nationality in Xinjiang.
