| Objective: To detect the levels of vasohibin-2(VASH-2)and F-box and WD-40 domain protein 7(F-box and WD-40 domain protein 7,FBXW7)in the tissues and serum of patients with lung adenocarcinoma To explore the changes in the expression levels of VASH-2 and FBXW7 in lung adenocarcinoma and their clinical significance.Methods: Randomly select 30 patients who were admitted to the Department of Thoracic Surgery of the First Affiliated Hospital of Hebei North University from April 2019 to April 2020 and were diagnosed with lung adenocarcinoma by the Department of Pathology.The lung cancer tissues and paracancerous tissues of the patients were collected((>5 cm away from cancer tissue)and serum,30 cases of healthy physical examination were collected at the same time.Collect clinical data of patients,use immunohistochemistry,fluorescence quantitative PCR,and enzyme-linked immunosorbent assay to detect the expression levels of VASH-2 and FBXW7 in each group,compare the differences in expression between VASH-2 and FBXW7 in each group,and analyze the differences between VASH-2 and FBXW7.Whether the expression of FBXW7 is related to different clinicopathological parameters;using SPSS22.0 statistical software,the immunohistochemical results are calculated by chi-square test to calculate the positive expression rate,and Spearman rank correlation method is used to analyze the relationship between VASH-2 protein and FBXW7 protein in lung adenocarcinoma Correlation;independent sample t-test was used to analyze the relationship between the expression of VASH-2 and FBXW7 and relevant clinicopathological characteristics of lung adenocarcinoma in q RT-PCR results;the area under the receiver operating characteristic curve(AUC)was used to assess VASH-2 in tissues and FBXW7 in the diagnosis of lung adenocarcinoma.Results: 1.The positive expression rate of VASH-2 in cancer tissues of lung adenocarcinoma patients was significantly higher than that in adjacent tissues,and the difference was statistically significant(P <0.05);the positive expression rate of FBXW7 in lung adenocarcinoma patients was significantly lower than that in cancer tissues The difference was statistically significant in adjacent tissues(P <0.05);the expression of VASH-2 in lung adenocarcinoma patients’ tissues and serum was significantly higher than that in the control group,and the difference was statistically significant(P <0.05);FBXW7 in lung glands The expression level in tissues and serum of cancer patients was significantly lower than that in the control group,and the difference was statistically significant(P<0.05);2.The expression of VASH-2 in lung adenocarcinoma patients and the degree of tumor differentiation,TNM staging and lymph node metastasis All related(P<0.05).The expression of VASH-2 in the tissues of lung adenocarcinoma patients has nothing to do with the gender and age of the patients(P>0.05);the expression of FBXW7 in the tissues of lung adenocarcinoma patients is related to the degree of tumor differentiation and TNM stage(all P<0.05).The expression of FBXW7 in the tissues of lung adenocarcinoma patients was not related to the gender,age,and the presence or absence of lymph node metastasis(P>0.05).3.Spearman rank correlation analysis immunohistochemical results:VASH-2 and FBXW7 expression in lung adenocarcinoma patients were negatively correlated,the difference was statistically significant r=-0.431(P<0.05).Pearson correlation analysis q RT-PCR results: VASH-2 showed a decreasing trend with the increase of FBXW7,the expression of the two was negatively correlated,and the correlation coefficient r was-0.752(P<0.05).The combined detection of VASH-2 and FBXW7 in the tissues of patients with lung adenocarcinoma has an AUC of 0.941(95% CI:0.882-1.000),and the sensitivity and specificity are 0.867 and 0.933,respectively.Conclusion: 1.VASH-2 is highly expressed in lung adenocarcinoma,which is related to tumor progression and metastasis;FBXW7 is lowly expressed,which is related to tumor progression.2.VASH-2 and FBXW7 may play a role in the progression of lung adenocarcinoma.3.The expression trends of VASH-2 and FBXW7 in lung adenocarcinoma tissue and serum of patients with lung adenocarcinoma are basically the same. |
