Objective:To investigate the effect of β-TCP/PLGA scaffold loaded with bone promoting drug HA15 on bone defect.Methods:(1)Construct the bone tissue scaffold loaded with HA15,and test its mechanical support strength and drug release ability.(2)Detect the expression of osteogenic related genes and proteins of mouse bone marrow mesenchymal stem cells treated with HA15.(3)Observe the effect of the bone tissue scaffold loaded with HA15 in rabbit radial bone defects.Results:(1)HA15 loaded β-TCP/PLGA bone tissue scaffold had the same biomechanical properties as cancellous bone and suitable drug release ability.(2)HA15 could target HSPA5 and promote the differentiation of mouse bone marrow mesenchymal stem cells into osteoblasts.(3)HA15 loaded β-TCP/PLGA bone tissue scaffold could promote bone regeneration and angiogenesis around bone defect in rabbitsConclusion:HA15 loaded β-TCP/PLGA bone tissue scaffold has good biomechanical properties and suitable drug release.HA15 can target HSPA5 and effect endoplasmic reticulum stress,thus promoting osteogenesis,bone regeneration and angiogenesis of rabbit bone defect model.
