| Objective: The calcium and phosphorus/ chitosan(Ca/P/CS)composite coating with good properties was prepared on the surface of ultrafine-grained titanium(UFG-Ti)to obtain better chitosan concentration parameters.Methods: The Ca/P/CS composite coating with different CS concentration(0.1 g/L,0.5g/L,1 g/L,2 g/L)was prepared by micro-arc oxidation(MAO)and dip-coating method.Mouse osteoblasts(MC3T3-E1)were cultured in vitro and inoculated on the surface of experimental(UFG-Ti based Ca/P/CS composite coating)and control(UFG-Ti based Ca/P composite coating)groups.The cytotoxicity,adhesion and proliferation of the osteoblasts were analyzed using the Cell Counting Kit-8(CCK-8)assay.The number and morphology of the cells were observed using a laser confocal microscope.The hemolysis properties were assessed using fresh rabbit blood and measured the optical density value for analysis.Results: The surface of UFG-Ti based Ca/P/CS composite coating treated by micro-arc oxidation and dip-coating presents a porous structure with uniform pore size and neat arrangement.Drug experiments have proved that chitosan has tendency to promote the growth of osteoblasts cells.The UFG-Ti based Ca/P/CS composite coating with different CS concentration was found to have no significant toxicity to osteoblasts,as well as the blood compatibility,was less than five per cent of the hemolytic standard.The adhesion and proliferation of osteoblasts in the Ca/P/CS composite coating with different CS concentration(0.1 g/L,0.5 g/L,1 g/L)on UFG-Ti based was better than that UFG-Ti based Ca/P composite coating group.The Ca/P/CS composite coating experimental group with different concentrations of CS showed that the 0.5 g/L group had a more obvious effect on cell adhesion and proliferation(P<0.05).There was no significant difference between the 2 g/L group and UFG-Ti based Ca/P composite coating group(P>0.05).Laser confocal microscopy showed that the morphology and quantity of cells in the experimental group with UFG-Ti based Ca/P/CS composite coating were more ideal.Conclusions: The UFG-Ti based Ca/P/CS composite coating with suitable chitosan concentration can promote the adhesion,proliferation and differentiation of osteoblasts.The 0.5g/L of UFG-Ti based Ca/P/CS composite coating has relatively better biological properties. |
