Objective:Observe the sciatic nerve function and the expression of S100,MBP,and p-p38 protein,the morphological changes of the sciatic nerve and the morphological changes of the myelin sheath of the sciatic nerve by observing the sciatic nerve squeezing injury model in rats.Aiming at the mechanism of axon regeneration and repair,it provides clinical experimental basis.Method:Eighty-one male SD rats were randomly divided into three groups: sham operation group,model group,and electroacupuncture group,with 27 rats in each group;according to the time of 3rd,7th,and 14th day after operation,they were divided into three subgroups,each subgroup Group of 9 animals.The model group and the electro-acupuncture group clamped the right sciatic nerve of the rat with hemostatic forceps,resulting in a crush injury model.In the sham operation group,only the right sciatic nerve was stripped without clamp.In the electroacupuncture group after the operation,the Huantiao and Weizhong acupoints were treated with electroacupuncture from the 1st day,once a day,20 minutes each time,for a total of 14 days.The model group and the sham operation group only performed the same as the electroacupuncture group Bound and restraint,without any intervention treatment.After 3rd,7th and 14th day`s interventions were completed,the sciatic nerve function index(SFI)was tested to observe the changes in the motor function of the sciatic nerve in rats.After the samples were taken,the sciatic nerve was stained with HE to observe the changes in the morphology and structure of the sciatic nerve;the microstructure changes of the myelin sheath were observed under the transmission electron microscopy;the expression of S100 protein,MBP protein and p-p38 protein at the sciatic nerve injury point were detected by immunohistochemical method.Result:1.Sciatic nerve function index: On the 3rd day after the operation,compared with the sham operation group,the SFI of the model group and the electroacupuncture group both decreased significantly,and there was a statistical difference(P<0.05);on the 7th day and 14th day after the operation,compared with the model group,the SFI value of the electroacupuncture group was significantly higher,and there was a statistical difference(P<0.05).There were statistical differences in the electroacupuncture group on the 3rd and 7th day,the 7th and 14th day after the operation(P<0.05).2.HE staining results: on the 3rd day of the model group,nerve fibers were broken,demyelinated,axons disintegrated,and a large number of cell infiltrations were seen;on the 7th day,nerve fibers were broken and structurally disordered,surrounded by cell infiltration,and individual Schwann cells proliferated.On the 14th day,the nerve fiber injury was slightly repaired;there was a small amount of Schwann cell proliferation,and some axons began to form.On the 3rd day of the electroacupuncture group,the cell structure was similar to that of the model group on the 3rd day,but on the7 th day,a large number of Schwann cell proliferation was seen,the arrangement was more disordered,and some fibroblasts were seen;Multiple axons are formed.3.Immunohistochemical results:(1)S100 protein:on the 3rd day after operation,compared with the sham operation group,the S100 average optical density value(AOD value)of the model group and the electroacupuncture group was significantly decreased,and there was a statistical difference(P<0.05);After the 7th and 14th day,compared with the model group,the S100 protein AOD value of the electroacupuncture group was significantly increased,and there was a statistical difference(P<0.05).The electroacupuncture group was compared with 3rd and 7th after surgery,and 7th and 14th day after surgery,There are statistical differences in them(P<0.05).(2)MBP protein:on the 3rd days after operation,compared with the sham operation group,the AOD value of MBP in the model group and the electroacupuncture group was significantly decreased,and there was a statistical difference(P<0.05);After the 7th and the 14th day,compared with the model group,the AOD value of the electroacupuncture group was significantly higher,and there was a statistical difference(P<0.05).The electroacupuncture group was compared with the 3rd and the 7th day after operation,and the 7th and 14th day after operation.There are statistical differences in them(P<0.05).(3)p-p38 protein: on the 3rd day after operation,compared with the sham operation group,the p-p38 protein AOD value of the model group and the electroacupuncture group increased significantly,and there was a statistical difference(P<0.05);the 7th day to the 14th day after operation,compared with the model group,the AOD value of p-p38 protein in the electroacupuncture group was significantly reduced,and there was a statistical difference(P<0.05).There were statistical differences in the electroacupuncture group,between the 3rd and the 7th day,the 7th and the 14th day after the operation(P<0.05).4.Transmission electron microscopy results:3 days after operation in the model group,the myelin structure was damaged with severe edema;there was a small amount of new myelin on the 7th day,but the thickness of the myelin was thin;on the 14th day,there were a small amount of proliferating cells and new myelin,at this time The thickness of the sheath is acceptable,accompanied by more serious edema;3 days after electroacupuncture,the myelin structure is damaged;7 days,there is more new myelin,and the thickness of the myelin is increased;14 days,there is a lot of new myelin,and the new myelin is changed Thick,relatively uniform structure,accompanied by slight edema.Conclusion:1.Electroacupuncture at "Huantiao Point" and "Weizhong Point" can promote the recovery of the motor function of the sciatic nerve in rats.2.Electroacupuncture can promote the restoration of nerve cell morphology and structure after sciatic nerve injury.3.Electroacupuncture can promote the expression of S100 and MBP protein,promote the proliferation of Schwann cells,and promote the regeneration of myelin sheath.4.Electroacupuncture can inhibit the expression of p-p38 protein after sciatic nerve injury.Electroacupuncture can promote the repair and regeneration of axons after sciatic nerve injury by inhibiting the p38 MAPK signaling pathway.
