| Object:To gain the epidemiological data of adenovirus,astrovirus and sapovirus in children with acute diarrhea in Chongqing.And to find out the genotypes of the three viruses by PCR or RT-PCR,so as to better guide the prevention of diarrhea.Methods:Fecal specimens were collected from children with acute diarrhea in the outpatient department of Children’s Hospital of Chongqing Medical University from 2017 to 2019,the clinical data was recorded meantime.Adenovirus was detected by PCR,astrovirus and sapovirus were detected by RT-PCR.The epidemiological data including detection rate,seasonality,distribution of the three viruses in different genders and age groups were analyzed.The positive samples were analyzed to determine the genotype.Phylogenetic trees were created by MEGA 7.0,with Kimura’s two-parameter method to calculate the genetic distance.Neighbor-joining method and boot-strap analysis with 1000 times were also used in the study.Result:1561 specimens were collected from 2017 to 2019.There were114 positive cases,among which 1 case was infected by adenovirus and astrovirus,1 case was infected by adenovirus and sapovirus,and 1 case was infected by the three viruses.The detection rates of adenovirus,astrovirus and sapovirus were 4.04%(63/1561),2.63%(41/1561)and0.90%(14/1561)respectively.Among the 1561 children,there were 922 males and 639 females,and there was no significant statistical difference between males and females for the infection of the three viruses(P>0.05).Adenovirus was detected in all age groups and children under 3years old accounted for 90.48%(57/63).The highest positive rate was8.42%(8/95)at the age of 25 to 36 months,and the lowest was 0.75%(3/399)at the age of 0 to 6 months.Adenovirus was detected throughout the year and with the highest positive rate of 7.61%(14/184)in November,followed by 6.49%(5/77)in May.Astrovirus was detected in all age groups and children under 3 years old accounted for 85.37%(35/41).The highest positive rate was 5.17%(3/58)at the age of 61 to 165 months.Astrovirus was most detected in March with the positive rate of8.70%(10/115),followed by April with the positive rate of 7.78%(7/90).Sapovirus was detected in children aged 7 to 60 months old and children under 3 years old accounted for 92.86%(13/14).The highest positive rate was 2.43%(10/411)at the age of 13 to 24 months.Sapovirus was detected only in May,July,September,October,November,and December,with the highest positive rate of 3.26%(6/184)in November.Phylogenetic tree analysis showed that there were 43 strains of adenovirus specie F,accounting for 68.3%(43/63)of the total adenovirus positive cases,including 41 strains of F41,2 strains of F40.Meantime 6strains of A31,5 strains of B3,1 strain of B7,1 strain of C1,1 strain of C2,3 strains of C5,and 2 strains of C6 were also detected in the study.Two genotypes of astroviruses were detected,including 36 strains of HAstr V-1and 5 strains of HAstr V-5.Two genotypes of sapovirus were detected,including 13 strains of GⅠ.1,1 strain of GⅠ.2.Conclusion: Adenovirus,astrovirus and sapovirus were the important pathogens causing acute diarrhea in children in Chongqing.There was no gender difference in the infection of the three viruses.Children under 3years old were the main victims.Adenovirus can be detected throughout the year without seasonal pattern.Astrovirus was prevalent in spring.For the three viruses,F41、HAstr V-1、GⅠ.1 were the most prevalent genotype respectively.Adenovirus 31 type could cause diarrhea too.Adenovirus species B and C were related to acute diarrhea in children. |
