| Objective:In order to ensure the safety of clinical application of Dalbergia cochinchinensis,the establishment of serum pharmacochemistry and quality standards of Dalbergia cochinchinensis was studied under the guidance of the 2020 edition “Chinese Pharmacopoeia” and serum pharmacochemistry of TCM.This study laid a foundation for further clarifying the effective basic substances in Dalbergia cochinchinensis.Methods:1.Based on the theoretical guidance of serum pharmacochemistry,UPLC-Q-TOF technology was used to study the serum pharmacochemistry of water extract from heartwood of D.cochinchinensis.The detection data were imported into Peakview software.the components in the plasma of the water extract of D.cochinchinensis and the rats after oral administration of it were analyzed and identified based on the mass spectrometry database of Dalbergia plant compounds which established by ourselves and MASSBANK online database.The prototype components and metabolites of D.cochinchinensis after oral administration were determined.2.The quality standard of D.cochinchinensis was studied based on the latest edition of "Chinese Pharmacopoeia".It is mainly divided into four parts.Firstly,the identification of D.cochinchinensis was studied,including powder identification,cross section identification and thin layer identification.Secongly,general inspection items,including moisture,ash,acid insoluble ash,extract content determination,heavy metal inspection studies.Again,study on fingerprint of Pterocarpus alternatus.Finally,determination of Liquiritigenin and Latifolin in D.cochinchinensis.Results:1.A total of 94 compounds were identified by positive and negative scanning modes,which could be divided into flavonoids,aromatic acids,new flavonoids,glycosides,benzofurans,etc.The main component was flavonoids.On this basis,guided by the theory of serum pharmacochemistry of traditional Chinese medicine,UPLCQ-TOF technology was used to detect and analyze the plasma of rats after oral administration of water extract of D.cochinchinensis heartwood.A total of 60 components were identified,including 24 prototype components which including flavonoids,aromatic acids,glycosides,terbinoids and phenols and 39 metabolic components.2.In this paper,the identification of D.cochinchinensis was studied to verify the sources of 10 batches samples in this experiment.In the study of thin-layer chromatography of D.cochinchinensis,the silica gel plate and its manufacturers,sample size and expansion conditions were investigated in this paper.Finally,Qingdao Haiyang silica gel G plate was used,with a sample size of 2 μL and expansion conditions of petroleum ether : chloroform : methanol : formic acid =10 : 50 : 2 : 1 as the TLC identification conditions of D.cochinchinensis.3.Based on the 2020 edition of “Chinese Pharmacopoeia”,this paper studied the general inspection items such as water content,ash content,acid insoluble ash content and the content of extract of D.cochinchinensis.Results the moisture content of 10 batches of samples was 7.51-11.98%,the ash content was 0.12-0.58%,and the acid insoluble ash content was 0.01-0.25%.In addition,this paper investigated the methods for the extraction of D.cochinchinensis,and finally,the content of 10 batches of D.cochinchinensis was 13.05%-31.45% by hot extraction and 70% ethanol.4.In the establishment of HPLC fingerprint method of D.cochinchinensis.the conditions were finally determined as follows: C18 column;column temperature:35 ℃;Detection wavelength: 254 nm;injection volume: 10 μL;The mobile phase was 0.03% phosphoric acid solution(A)-acetonitrile(B);Gradient elution: 0-10 min,15%-25% B;10-25 min,25%-30% B;25-60 min,30%-50% B;60-75 min,50%-70% B.When the contents of Liquiritigenin and Latifolin in D.cochinchinensis were detected,the contents of Liquiritigenin and Latifolin were0.81-3.92 mg/g and 19.17-49.57 mg/g,respectively.Conclusion:The chemical composition of D.cochinchinensis heartwood is complex,mainly flavonoids,followed by organic acids,new flavonoids,glycosides,coumarins,benzofurans and so on.In the study of its serum pharmacochemistry,it was found that the flavonoids in D.cochinchinensis were mainly introduced into the blood by prototype components and played a role.In addition,the method of TLC identification,HPLC fingerprint and content determination of D.cochinchinensis was established by studying the quality standard of D.cochinchinensis.The method is accurate,feasible and stable,which can provide systematic evaluation for the quality control of D.cochinchinensis. |
