In this paper,the processing integration technology of Chaenomeles speciosa(C.speciose)in producing area was studied,and the quality of fresh cut Chaenomeles speciosa(processing integration technology in producing area)was compared with that of traditional processing technology.The quality standard of fresh cut Chaenomeles speciosa was drawn up,which opened up a new way for Chaenomeles speciosa industrial processing.A method for simultaneous determination of five pentacyclic triterpenoids(3-O-acetylpalmitol acid,betulinic acid,oleanolic acid,ursolic acid and 3-O-acetylursolic acid)in Chaenomeles by high performance liquid chromatography(HPLC)was established.QAMS of eight batches of Chaenomeles from four producing areas in China,which were processed by fresh cut technology and traditional technology,were determined by HPLC In order to provide a new analytical model for multi index quality control standard of Chaenomeles speciosa.Objective:To conduct a research on the integration of processing in producing area,compare the quality of C.speciose slices processed by fresh cut technology and traditional technology,demonstrate the feasibility of integrated processing technology applied to C.speciose,and establish the processing standard of integrated processing of fresh-cut slices;establish the HPLC method for determination of pentacyclic triterpenoids in C.speciose;apply the method of HPLC to the determination of pentacyclic triterpenoids in C.speciose and evaluate the quality of fresh-cut and traditional C.speciose slices.Method:1.Investigate the influence of different cutting methods,drying methods and drying temperature in the fresh-cut process on the intrinsic quality of fresh-cut decoction pieces(the content of oleanolic acid and ursolic acid and the drying yield)to determine the integrated processing technology(fresh Cutting process).2.Eight batches of C.speciose samples were collected from four different producing areas in China,which were processed by fresh cut technology and traditional technology respectively.According to the 2020 edition of Chinese Pharmacopoeia,the properties of each batch of C.speciose were observed,and the water content,total ash content,acidity,alcohol soluble extract,total content of oleanolic acid and ursolic acid in C.speciose were determined.Meanwhile,the content of chlorogenic acid was also determined.Finally,the quality standard of fresh cut C.speciose slices was formulated.Chromatographic conditions for determination of chlorogenic acid content: Chromatographic column: COSMOSⅡ 5C18-MS-Ⅱ(250×4.6mm,5μm).The mobile phase was acetonitrile-0.4% phosphoric acid solution(13:87);the injection volume was10.0μL,the flow rate was 1.0m L/min,and the detection wavelength was 327 nm.3.Using extraction temperature,ethanol concentration,liquid-to-material ratio,and extraction time as independent variables,liquid-phase analysis of oleanolic acid in the C.speciose extracted by ultrasound to obtain the concentration of oleanolic acid in the extract is the dependent variable.Carry out Box-Behnken response surface analysis to optimize the extraction method of C.speciose.4.The chromatographic conditions of oleanolic acid and ursolic acid in Pharmacopoeia were optimized by changing the composition of mobile phase.The optimized chromatographic conditions were used to analyze the C.speciose extract(extracted by the optimized extraction method),confirmthe components to be determined by QAMs.The unknown compounds needed to be determined by atmospheric pressure silica gel column chromatography,thin layer chromatography and Sephadex LH-20,the preparation of liquid equivalent materials and methods to separate and identify,and finally a HPLC method for the determination of pentacyclic triterpene in C.speciose was established.5.Eight batches of C.speciose samples collected from four producing areas in China were processed by fresh cut and traditional techniques respectively.The quality of the eight batches of C.speciose samples was evaluated by QAMS.Result:1.the integrated processing technology of C.speciose production area(fresh cut C.speciose slices processing process)is: cut 2-4 pieces of fresh C.speciose vertically,then slice the slice,the thickness of the slice is 2-4mm,and the temperature is set to 80 ℃ for hot air drying for6-7 hours.2.eight samples of C.speciose were processed by fresh cutting and traditional adding technology.The characters of traditional C.speciose pieces meet the requirements of Pharmacopoeia,and the differences between fresh cut C.speciose and traditional C.speciose slices are obvious.The total content of water,ash,acidity,alcohol soluble extract,oleanolic acid and ursolic acid in C.speciose of 8 batches of samples all conform to the requirements of Chinese Pharmacopoeia 2015,and the content of chlorogenic acid is greater than 0.1%3.Box-Behnken response surface method was used to optimize the extraction process of oleanolic acid from C.speciose.The optimal extraction conditions were as follows: extraction temperature 50 ℃,ethanol concentration 90%,liquid-solid ratio 30:1,extraction time 40 min.4.The chromatographic conditions of the method are as follows: acetonitrile-0.005 mol / L diamine hydrogen phosphate solution(adjust p H to 6.5 with phosphoric acid)(70 ∶ 30)as mobile phase;flow rate is 1 m L/ min;detection wavelength is 210 nm;column temperature is30℃;injection volume is μL;Five pentacyclic triterpenoids were identified as 3-O-acetyl pomonolic acid,betulinic acid,oleanolic acid,ursolic acid and 3-O-acetyl ursolic acid.5.The content of oleanolic acid and ursolic acid in fresh cut C.speciose from the same place was 5.60% higher than that in traditional C.speciose,and the content of five pentacyclic triterpenoid acids was 10.92% higher than that in traditional C.speciose.Conclusion:Compared with the traditional processing technology,the integration of producing area processing of C.speciose reduces the processing procedures and transportation costs,reduces the waste of manpower and energy,The integrated processing technology of C.speciose and the established processing standard of fresh cut C.speciose slices provide support for the better and more standardized development and utilization of C.speciose resources.QAMS established in this study realizes the multi index quality evaluation of C.speciose,and helps to improve the quality control standard of C.speciose.The quality evaluation of five pentacyclic triterpenoids in C.speciose by QAMS can control the quality
