| Background:Instability of M1(pro-inflammatory)/M2(anti-inflammatory)macrophage ratio is closely related to atherosclerosis,artificial regulation of macrophage polarization to M2 phenotype is expected to achieve anti-atherosclerosis therapy.Animal studies have shown that GLP-1R may be involved in macrophage polarization.However,the distribution of GLP-1R on the surface of human macrophages and whether GLP-1R is involved in mediating macrophage polarization and thus plays a role in atherosclerosis remain unclear.Objective:To investigate the expression of GLP-1R and cytokines in macrophages of CHD and their correlation with blood lipid,as well as the effect of GLP-1R on the differentiation of macrophages in the process of atherosclerosis.Methods:Totally,49 patients with coronary heart disease(CHD)and 52 cases of health control(HC)were recruited,all subjects accept coronary angiography gold standard inspection.The conditions of CHD group was defined as no less than one left main artery(LM),left anterior descending branch(LAD),left circumflex branch(LCx)or right coronary artery(RCA)stenosis≥50%,and those with stenosis less than 50%(including no obvious stenosis)were assigned to the control group.Flow cytometry were used to detect the percentage of M(CD14~+),M1(CD14~+CD80~+),M2(CD14~+CD206~+)macrophages and their surface GLP-1R expression differences in the two groups.Using BD cytokine kit to detect the levels of IL-8,IL-1β,IL-6,IL-10,TNF,IL-12p70 in plasma.Results:GLP-1R expression on the total and M2 macrophages membrance was lower in the CHD group and the difference was statistically significant(P<0.05),while the level of IL-8 was higher in the control group(P<0.05).No significant differences in the percentage of total macrophages,M1 and M2 macrophages,the expression level of GLP-1R on M1 macrophages or the concentration of other 5cytokines between the two groups.After adjusting for potential confounders with multivariate linear regression,the decrease in Gensini score was accompanied by an increase in the percentage of M1 macrophages.Conclusion:GLP-1R agonist is independent of the hypoglycemic effect of T2DM and has protective effect on cardiovascular system.GLP-1R may regulate the polarization of macrophages toward M2,thus playing a protective role in the progression of coronary atherosclerosis. |
