| Objective:1.To analyze the drug sensitivity of clinical isolates of Neisseria gonorrhoeae to seven antibiotics(ceftriaxone,cefixime,spectinomycin,azithromycin,tetracycline,ciprofloxacin,penicillin)in Taiyuan,Shanxi Province.2.To analyze the relationship between sensitivity of Neisseria gonorrhoeae to ceftriaxone and the mutations of drug resistance genes penA and ponA in Taiyuan,Shanxi Province.Method:1.From January 2019 to December 2020,specimens of genitourinary tract secretions suspected of Neisseria gonorrhoeae infection in the Department of Dermatology and Venereology of the second hospital of Shanxi Medical University were collected for isolation,culture and identification;The susceptibility of Neisseria gonorrhoeae to seven antibiotics(ceftriaxone,cefixime,spectinomycin,azithromycin,tetracycline,ciprofloxacin,penicillin)was detected by agar dilution method.2.The DNA was extracted from clinical isolates of Neisseria gonorrhoeae.Gene fragments of Pena and PONA were amplified and sequenced;The gene sequences of Neisseria gonorrhoeae were compared with those of NCBI,and the relationship between amino acid substitution and susceptibility of Neisseria gonorrhoeae to ceftriaxone was analyzed.Result:1.45 strains of Neisseria gonorrhoeae were not resistant to ceftriaxone,cefixime and spectinomycin.The drug resistance rates of azithromycin,tetracycline,ciprofloxacin and penicillin were 15.55%,100.00%,97.78% and 100.00% respectively,among which TPNG was 22.22%;PPNG was 51.11%;The low sensitivity rates of Neisseria gonorrhoeae to ceftriaxone,cefixime and azithromycin were 6.67%,11.11% and 17.78%.2.Among the 45 strains,14 PBP2 patterns were found,including 13 non mosaic patterns and one mosaic pattern(XXXIV,n = 3);Among 45 clinical isolates of Neisseria gonorrhoeae,40 strains showed L421 P mutation in ponA.Conclusion:1.Azithromycin,penicillin,tetracycline and ciprofloxacin are not suitable for the treatment of Neisseria gonorrhoeae in Taiyuan area,but ceftriaxone,cefixime and spectinomycin can still be used for the treatment of Neisseria gonorrhoeae in this area.2.Specific non mosaic penA mutation patterns(XVII and XIII)may play an important role in the reduction of the sensitivity of Neisseria gonorrhoeae to ceftriaxone in this region.The non mosaic XXXIV may not be related to the decrease of sensitivity of Neisseria gonorrhoeae to ceftriaxone in this region.(2)The substitution of L421 P in ponA gene may have little effect on the susceptibility of Neisseria gonorrhoeae to ceftriaxone. |
