Research On Association Between PAD4 And Deep Vein Thrombosis

Objective:To compare the level of PAD4(Peptidylarginine deiminase 4,PAD4)in peripheral venous blood between the patients diagnosed as lower extremity deep vein thrombosis(DVT)and healthy people,explore the correlation between PAD4 and DVT of lower extremity,and investigate its diagnostic value for DVT.Hope to provide new directions for diagnosis and treatment of DVT。Methods:A case-control study was adopted to enroll 50 inpatients with DVT in the Department of Vascular Surgery of Shanxi Bethune Hospital and 50 healthy examiners in the physical examination center.After admission,the patients’ fasting peripheral venous blood was collected in the morning,and the serum PAD4 was detected by enzyme-linked immunosorbent assay.Comparing the levels of PAD4 between the two groups of patients,analyze the correlation between the level of PAD4 in DVT patients and their blood cells and coagulation indicators,thrombotic limbs,thrombosis type,and whether it is combined with pulmonary embolism;drawing ROC curve to make it clear whether it has diagnostic value for DVT,look for its diagnostic cut point and diagnostic value。Results:1.A total of 50 DVT patients and 50 healthy subjects were included in the study.There difference of age,gender and body mass index(BMI)between the two groups was not statistically significant;2.Compared with healthy subjects,the level of PAD4 in peripheral blood serum of patients with DVT was significantly increased(1.385 [0.862-2.347] vs 0.677[0.458-1.103],p<0.01).Binary Logistic analysis showed that PAD4 is an independent risk factor for lower limb DVT(OR=4.52,95% CI: 2.03-10.06,p<0.01).3.In patients with DVT,the level of peripheral blood serum PAD4 has no correlation with blood cells,coagulation indicators,thrombotic limbs,thrombosis type and whether it is complicated with pulmonary embolism.4.Further analysis of ROC curve suggested that the content of PAD4 has diagnostic value for DVT patients(AUC: 0.794,p<0.01),the Cut-off value is 1.207 ng/ml,the sensitivity is 62% and the specificity is 84%.Conclusion:The level of PAD4 in peripheral venous blood of patients with DVT is significantly higher than that in healthy subjects.It is an independent risk factor for DVT in lower limbs.ROC curve suggests that it has diagnostic value.PAD4 may be used as a new target for the diagnosis and treatment of DVT in the future.
