Objective:To study the expression of Dickkopf-related protein 3(DKK3)gene in cervical squamous cell carcinoma(CSCC)tissues,high-grade cervical squamous intraepithelial lesion(HSIL)tissues,low-grade cervical squamous intraepithelial lesion(LSIL)tissues and normal cervical tissues,and to compare the differences in their expression levels,and to analyze the relationship between the expression level of DKK3 and the grade of cervical lesions.Methods:Patients were selected as the research objects,who underwent colposcopy and cervical biopsy in the Department of Obstetrics and Gynecology of the Second Hospital of Shanxi Medical University from July 2020 to October 2020.There were 30 patients in the normal cervical group,30 patients in the low-grade cervical squamous intraepithelial lesion group,30 patients in the high-grade cervical squamous intraepithelial lesion group and 30 patients in the cervical squamous cell carcinoma group.All patients were confirmed by the results of cervical histopathological examination.The study specimens were collected from the above-mentioned subjects,all of which were taken at the same time during cervical biopsy,and cryopreserved in-80℃ refrigerator for a long time.Quantitative real-time PCR(q RT-PCR)assay was used to detect the relative expression level of DKK3 m RNA in four types of tissues;the expression of DKK3 protein was examined by Western blotting in four types of tissues.SPSS 22.0 statistical software was used to analyze the data.For measurement data with normal distribution and uniform variance,one-factor analysis of variance was used for comparisons between groups,and LSD-t test was used for multiple comparisons between groups,with the test level α=0.05.Results:1.Compared with the normal cervical group,the expression of DKK3 m RNA was lower in the CSCC group,HSIL group and LSIL group(p < 0.001,p < 0.001,p = 0.038).The expression of DKK3 m RNA in the CSCC group and HSIL group was lower than that in the LSIL group(p < 0.001,p = 0.006).The expression of DKK3 m RNA in the CSCC group was lower than that in the HSIL group(p = 0.049).2.Compared with the normal cervical group,the expression of DKK3 protein was lower in the CSCC group,HSIL group and LSIL group(p<0.001,p<0.001,p<0.001).The expression of DKK3 protein in the CSCC group and HSIL group was lower than that in the LSIL group(p < 0.001,p = 0.005).The expression of DKK3 protein in the CSCC group was lower than that in the HSIL group(p < 0.001).Conclusion:The expression of DKK3 was decreased in the cervical squamous intraepithelial lesion tissues and cervical squamous cell carcinoma tissues.The expression of DKK3 was higher in the cervical squamous intraepithelial lesion tissues than that in the cervical squamous cell carcinoma tissues.The expression of DKK3 in the low-grade cervical squamous intraepithelial lesion tissues was higher than that in the high-grade cervical squamous intraepithelial lesion tissues. |
