Objective:To study the chemical constituents from the dried rhizome of Menispermum dauricum DC.especially the alkaloids.To study its anti-ACh E and anti-tumor activity.Methods:From the perspective of natural medicinal chemistry,this experiment uses the classical reflux extraction method to extract the dried rhizomes of Menispermun dauricum with 95%ethanol.The solubility of alkaloids in different p H solutions is used to extract alkaloids from their alcohol extracts.The total alkaloids are extracted from organic solutions of different polarities,and then separated and purified by various chromatographic separation methods of modern natural products,and the physical and chemical properties of the separated monomer compounds are compared.Then through various spectroscopic methods for structural identification;Galantamine was used as a control to use ELLMAN method to perform in vitro acetylcholinesterase activity experiments on some compounds;MTT method was used to preliminarily test the anti-tumor cytotoxic activity of some compounds.Results:In this study,17 alkaloid compounds were purified from the dried rhizomes of Menispermum dauricum,identified as:Pecrassipine B(1),Menidaurine A(2),Thalifoline(3),Acutumine(4),O-methylcorypalline(5),sinomenine(6),armepavine(7),Daurisoline(8),Dauricine(9),Pycnarrhine(10),O-methyldauricine(11),Amurolin(12),N-methylcorydaldine(13),Corypalline(14),Dauricumidine(15),(+)-magnoflorine(16),(R)-6-Methoxy-1-(4-methoxybenzyl)-2-methyl-1,2,3,4-tetrahydroisoquinolin-7-ol(17);some alkaloids have a certain inhibitory activity on acetylcholinesterase,with IC50values ranging from 25.8-64.8μM.The tested alkaloids showed certain anti-tumor activity,with IC50values ranging from 20.0-68.5μM.Conclusion:A total of seven structural types of alkaloids were isolated from the rhizomes of Menispermun dauricum:Isoquinoline(Compound 10);Dibenzylisoquinoline alkaloid(compounds 8,9,and 11);Monobenzyl isoquinoline alkaloid(Compounds 1,2,7,17);Tetrahydroisoquinoline alkaloid(compounds 5,14);Isoquinolinone alkaloids(Compounds3,13);Morphanes alkaloids(compounds 4,6,15);Aporphinoid alkaloids(compounds12,16).In addition,compounds 10 and 12 are isolated from this plant for the first time.The isolated alkaloids of the seven structures all have inhibitory activity on Ach E.The results show that the seven structural types of alkaloids all have a certain degree of anti-tumor activity.
