Objective:Explore the expression of signal transducer and activator of transcription STAT5 A and STAT5 B in T-cell lymphoblastic lymphoma(T-LBL),Analyze the relationship between STAT5 A and STAT5 B and the occurrence,development,and survival prognosis from the m RNA and protein levels,and provide new potential targets for the clinical diagnosis and treatment of T lymphoblastic lymphoma.Methods:(1)Use immunohistochemistry(IHC)methods to examine the expression of STAT5 A and STAT5 B in T-LBL tissues,and set up an experimental group and a control group respectively.The experimental group consisted of 120 T-LBL patients archived wax blocks with complete clinicopathological data and follow-up records in the Department of Pathology of Shanxi Cancer Hospital from 2001 to 2020.The control group consisted of 60 randomly selected archived paraffin specimens of lymph node reactive hyperplasia(LH).Immune response integration(IRS)was used to interpret the results,and the differences in STAT5 A and STAT5 B protein expression between the experimental group and the control group were compared.(2)Using quantitative real-time polymerase chain reaction(qRT-PCR)method to detect the expression of STAT5 A m RNA and STAT5 B m RNA in 60 T-LBL paraffin tissue specimens and 30 LH paraffin tissue specimens Happening.(3)Use SPSS 26.0 statistical analysis software to analyze the protein and m RNA expression of STAT5 A and STAT5 B in T-LBL and LH tissues,and Analyze its relationship with the clinicopathology and survival prognosis of patients with T-LBL.Results:(1)Interpretation of IHC staining results of STAT5 A and STAT5BAccording to the Immune Response Scoring(IRS),The staining results of STAT5 A and STAT5 B were interpreted as the product of the positive cell ratio score and the staining intensity score.Among 120 T-LBL cases,The positive expression rate of STAT5 A protein was 67.5%(81/120),of which 10 cases were weakly positive(+),59 cases were positive(++),12 cases were strongly positive(+++),and the protein was stained in the nucleus;Among the 60 reactive hyperplastic lymph node tissue samples in the control group,STAT5 A protein was mainly weakly positive or not expressed,and the positive expression rate was 5%(3/60).STAT5 B protein was strongly positively expressed,and the positive protein expression rate was 72.5%(87/120),of which 7 cases were weakly positive(+),10 cases were positive(++),and 70 cases were strongly positive(+++).The protein is stained in the cytoplasm and nucleus;In the 60 samples of reactive hyperplastic lymph node tissues in the control group,the STAT5 B protein was mainly weakly positive or not expressed in the germinal center,and the positive expression rate was 20%(12/60).The difference in the positive expression rate of STAT5 A and STAT5 B protein in T-LBL and LH was statistically significant(P<0.05).(2)The relationship between the expression of STAT5 A and STAT5 B and the clinicopathological characteristics of T-LBL patientsThe expression of STAT5 A protein was related to the patient’s gender(P<0.05),but not related to the patient’s age,IPI score,Ann Arbor staging,presence or absence of B symptoms,or complication of bone marrow.The expression of STAT5 B protein is related to the patient’s age and the presence or absence of B symptoms(P<0.05),and has nothing to do with gender,IPI score,Ann Arbor staging,or complication of bone marrow.And 61 cases were positive for both STAT5 A protein and STAT5 B protein(STAT5A+/STAT5B+).Combined with the analysis of various clinicopathological parameters,the STAT5A+/STAT5B+ group was positively correlated with the patient’s Ann Arbor staging(P<0.05),but there was no significant correlation with the patient’s gender,age,mediastinal widening,LDH level,and bone marrow complication.(3)The relationship between the expression of STAT5 A and STAT5 B protein and the survival time of T-LBL patientsIn this experiment,the deadline for follow-up of 120 T-LBL patients is August 30,2020,of which 93 have died and 27 have survived.The overall survival of patients with high expression of STAT5 A and STAT5 B protein was lower than those with low expression,and the difference was statistically significant(P<0.05).The overall survival of STAT5A+/STAT5B+ patients was significantly lower than that of other patients,followed by STAT5A-/STAT5B+ patients.(4)qRT-PCR resultsAmong the 60 T-LBL paraffin tissues in the experimental group,the relative expression of 30 LH tissues was used as the mean value.The relative expression of STAT5 A m RNA showed an up-regulated trend,and the difference was statistically significant(P<0.05).However,the relative expression of STAT5 AB m RNA in the T-LBL group was only 51.7% higher than the average,the expression was not significantly up-regulated,and the difference was not statistically significant(P>0.05).Conclusion:(1)The results of immunohistochemistry showed that the expressions of STAT5 A and STAT5 B in T-LBL were significantly higher than those of control group LH.It shows that STAT5 A and STAT5 B are closely related to the occurrence,development,infiltration and metastasis of T-LBL.Moreover,the expression of STAT5 A and STAT5 B is closely related to the gender of T-LBL patients and the presence or absence of B symptoms,which all indicate that the expression of STAT5 A and STAT5 B plays an important role in the development of T-LBL.(2)The results of qRT-PCR showed that the relative m RNA expression of STAT5 A and STAT5 B in T-LBL had different upward-regulation trends,but the protein expression levels of the two were not completely consistent with the m RNA expression levels.A deeper level requires further research and discovery.
