Objective:To analyze the relationship between Fibrinogen-like Protein 2(FGL2)and gliomas and explore the potential prognostic value and the immunotherapeutic targetability of FGL2 in glioma using a bioinformatics analysis.Methods:TCGA,GETx and GEO databases were used to analyze the transcriptional level of FGL2 in glioma tissues.Clinical data of low-grade glioma and glioblastoma(GBM)patients from TCGA database were used to analyze the relationship between FGL2 expression and clinicopathological characteristics.Data were analyzed using Kaplan-Meier survival analysis and univariate and multivariate Cox regression analyses.Gene set enrichment analysis(GSEA)was used to screen signal transduction pathways of FGL2.In addition,we used CIBERSORT,an established algorithm,to compare the differences in immune cells between samples with different FGL2 expression levels and evaluate the differences in the proportion of 22 immune cells in each sample.Gene Set Variation Analysis(GSVA)and correlation analysis were used to investigate the relationship between FGL2,infiltrated inflammatory immune cells,and immune checkpoint molecules.Results:FGL2 was highly expressed in gliomas,and its high expression was related to poor prognosis and progression of clinicopathological characteristics.GSEA showed that the high FGL2 phenotype was related to some immune-related pathways.GSVA and correlation analysis found that the expression of FGL2 was closely related to inflammatory response and immune response,and the degree of immune cell infiltration was high.Conclusion:FGL2 may be a potential prognostic biomarker and immunotherapy target for glioma.
