Background: Ischemic stroke has brought a huge burden to the health and economy of people all over the world.When ischemic stroke occurs,microglia can engulf damaged and necrotic neuronal cells and surrounding myelin,and at the same time polarize into different subgroups(M1,M2)at different times during the injury process.The role of M1 type microglia is to promote the occurrence of inflammation,and the opposite is M2 type microglia,which has anti-inflammatory effects.However,whether vinpocetine can affect the polarization of microglia during ischemic stroke to achieve the purpose of brain protection,this process has not yet been clear.Purposes: This experiment investigates the effect of vinpocetine on microglia polarization after ischemic stroke,and explores how vinpocetine affects the treatment and recovery of ischemic stroke,so as to provide a new theoretical basis for the mechanism of vinpocetine in the treatment of ischemic stroke.Methods: The experimental animals were C57BL/6 male mice,using transient middle cerebral artery occlusion(tMCAO)model,and they were divided into sham operation group,tMCAO+vehicle group,and tMCAO+vinpocetine group.The tMCAO+vinpocetine group was intraperitoneally injected with 10 mg/kg of vinpocetine,and the tMCAO+vehicle group was intraperitoneally injected with an equal volume of normal saline.After 3 consecutive injections,the material was perfused,TTC staining,immunofluorescence staining,qPCR,WB and other experimental methods were used to identify changes in cerebral infarction volume,activation of astrocytes,apoptosis of neurons,and polarization of microglia.Results: The results of TTC staining and neurological function score showed that the vinpocetine could significantly reduce the cerebral infarct volume of tMCAO mice and improve neurological function.The results of immunofluorescence staining showed that the number of apoptotic neurons in the infarct border and the number of activated astrocytes and microglia decreased in the tMCAO+vinpocetine group compared with the tMCAO+vehicle group;The WB experiment found that the expression of Bcl-2 protein in the tMCAO+vinpocetine group was higher than that in the tMCAO+vehicle group;while the protein expression of Bax and Cleaved Caspase-3 decreased;The protein expression levels of M1 microglia markers iNOS and CD16 decreased;the protein expression levels of M2 microglial markers ARG and CD206 increased.It was further found by qPCR that the m RNA expression levels of M1 microglia markers(CD16,CD32,CD11 b,iNOS)of different days in the tMCAO+vinpocetine group were lower than those in the tMCAO+vehicle group;while the M2 microglia markers(CD206,TGF-β,YM1/2,Arg 1)mRNA expression levels were higher than the tMCAO+vehicle group.Conclusion: Vinpocetine can reduce the infarct volume after ischemic stroke,inhibit neuronal apoptosis,and improve neurological function.Vinpocetine can inhibit the activation of glial cells after ischemic stroke,promote the polarization of M2 type microglia and inhibit the polarization of M1 type microglia,so as to achieve the effect of brain protection.
