Research background: Preeclampsia(PE)is one of the diseases with the highest incidence during pregnancy.Preeclampsia poses a serious threat to the health and safety of pregnant women and fetuses.In poor countries such as Africa and Latin America and Southeast Asia,millions of women have died of the disease.Because of the serious harm of preeclampsia,research on the disease is particularly important.Although there are many studies focusing on preeclampsia,the specific mechanism of the disease has not yet reached a consensus in the academic community.The serious disclosure of the etiology of preeclampsia still needs to be deepened.Preeclampsia is a placental-derived disease,so revealing the regulatory molecular mechanisms of placental development and function is self-evident for us to understand the causes of preeclampsia.Various mechanisms of action have been discovered by experts and scholars at home and abroad.In our previous work,we also found that a specific miRNA is significantly down-regulated in the placenta of patients with preeclampsia.This miRNA is miR-135b-5p.Purpose: Based on existing research and bioinformatics analysis,we speculate that miR-135b-5p may play an important role in the pathogenesis of preeclampsia by regulating the function of trophoblast cells.This study will be further applied in a larger sample population.To explore the correlation between miR-135b-5p and preeclampsia,and to reveal the expression characteristics of miR-135b-5p at the maternal-fetal interface and the relative expression abundance of trophoblast cells in different functional subgroups,for the next in-depth The study of the cellular biological pathways and molecular mechanisms of miR-135b-5p in preeclampsia points out the direction.Methods: The collection of obstetric hospitalizations in the Clinical College of Shenzhen Second Hospital of Anhui Medical University(Shenzhen Second People’s Hospital)and Shenzhen Longhua District People’s Hospital from January 1,2020 to August 31,2020 were in line with the diagnostic criteria of the ninth edition of the textbook 30 cases of pre-epilepsy pregnant women with cesarean section were selected as the research object(PE group).Another 30 cases of pregnant women who were given regular cesarean section in our hospital during the same period were selected as the normal pregnancy group(Normal control,NC group)..Collect placental tissue,label miR-135b-5p with RNAscope technology,observe the signal source under a microscope,and detect signal abundance for quantitative analysis.Results:(1)Location analysis: The expression abundance of miR-135b-5p is significantly different in different villi trophoblast cells.miR-135b-5p is expressed in high abundance in the villous syncytial trophoblast(STB),is also expressed in high abundance in the syncytial nodules(SK),and is moderately expressed in the infiltrating extravillous trophoblast cells(EVT)The expression abundance is expressed in low abundance in the chorionic trophoblast(CTB),but almost not expressed in the mesenchymal cells(MC)in the villi.(2)Semi-quantitative analysis: The expression of miR-135b-5p can be detected in the placenta of either the PE group or the NC group.The signal intensity results showed that the relative expression of miR-135b-5p in the preeclampsia group(18.1±3.9)was significantly lower than that of the normal control group(25.1±3.9),and the difference was statistically significant.Conclusion:(1)miR-135b-5p is mainly distributed in the villi syncytiotrophoblast and syncytial nodules,less in the cytotrophoblast,and almost not expressed in mesenchymal cells.(2)In PE patients,the expression of miR-135b-5p was significantly down-regulated,indicating that miR-135b-5p may be related to PE,and the specific relationship path needs to be further studied.(3)The relative expression abundance of miR-135b-5p in different functional subgroups of trophoblast cells indicates that this molecule may be involved in preeclampsia by regulating the structure and function of villi STB,or by regulating the invasion and migration ability of EVT.The process occurs.
