Saxifraga plant are perennial herb,has good medicinal properties.There are about400 species in the world,of which 203 are in China.They grow in both north and south of China,mainly in the high mountains of Qinghai,Gansu and other provinces.Tibetan medicine is mainly used to treat Hepatobiliary diseases.It is often used to reduce hepatochlic hygropyrexia,healing sore and rlieving pain.Studies have found that Saxifraga has a rich variety of chemical components and has good pharmacological activities.It has significant effects in antibacterial and antiviral,antioxidant,anti-tumor,anti-male and female sex hormones.Plants of this genus contain a variety of flavonoids,terpenoids,essential oils,polyphenols,glycosides,organic acids and others.Saxifraga umbellulata Hook.f.et Thoms.is a common Tibetan,which has been used as a traditional folk medicine for the treatment of hepatitis cholecystitis,infuenza,fever,sore heat,and so on.This thesis deals with the investigation on the chemical constituents and biological activity of Saxifraga umbellulata Hook.f.et Thoms.Systematic separation and purification of the petroleum ether extraction part and n-butanol extraction part of Saxifraga umbellulata Hook.f.et Thoms.Through modern spectroscopy technology and reference literature,a total of 15 compounds have been identified.The structures are:β-sitosterol(1),Emodin(2),dibutyl phthalate(3),hydroxyphenyl butanone(4),Hydroxydihydrobovolide(5-hydroxyl-3,4-dimethy-5-penty-l-2(5H)-furanone)(5),Tritetradecanol(6),Di(octylthio)methane(7),Undecanoic acid(8),8,10,13-Trimethyltetradecanoic acid(2E)-3-methylhexadec-2-enyl ester(tomentosate)(9),trans-9-octadecenoic acid methyl ester(10),3,7-dimethylnonadec-9-ene(11),isorhamnetin(12),3,5,7-trihydroxy-3′,4′-dimethoxyflavonoids(13),1,7-bis(3,4-dihydroxyphenyl)hepta-4E,6E-dien-3-one(14),(E)-1-(4-hydroxyphenyl)-7-(4-hydroxy-3-methoxyphenyl)hept-1-en-3-one(15).The antibacterial activity,antioxidant activity and anti-inflammatory activity of the four extracts were tested.By measuring the inhibitory ability of the four extracts on the five tested bacteria,the results show that the extracts of each polar segment of Saxifraga umbellulata Hook.f.et Thoms have a certain inhibitory effect on the five tested bacteria.Among them,the ethyl acetate part has a certain inhibitory effect on the five tested bacteria.The extract has the best inhibitory effect on the five tested bacteria,and the MIC values are 1.5 mg/ml,2 mg/ml,3 mg/ml,1 mg/ml,2 mg/ml,respectively.By measuring the scavenging ability and total reducing power of the four extracts on DPPH free radicals and hydroxyl free radicals,it is shown that each polar segment has a certain scavenging ability on free radicals,and all have a certain reducing ability,but their activities are all weak.In the positive control Vc.Among them,the IC50 of the ethyl acetate extract for scavenging DPPH free radicals was 0.67 mg/m L,and the IC50for scavenging hydroxyl free radicals was 0.44 mg/m L,and the ethyl acetate extract had a strong reducing ability.Compared with several other polar parts,the ethyl acetate extraction part has stronger antioxidant activity.The anti-inflammatory activity test results show that the four extracts have a significant inhibitory effect on the release of NO from mouse macrophages RAW 264.7 induced by LPS.Among them,the n-butanol extract has the strongest anti-inflammatory effect,with an inhibition rate of 68%and an IC50 of 0.45±0.14μg/m L.The isolated monomer compounds were studied for anti-inflammatory,anti-tumor andβ-lactamase inhibitory activity The anti-inflammatory activity of the monomer compounds were screened,and the results showed that none of the isolated monomer compounds had anti-inflammatory activity.The isolated monomer compounds were screened for anti-tumor activity.When the sample concentration was 100μg/m L,the compound Hydroxydihydrobovolide(5-hydroxyl-3,4-dimethy-5-penty-l-2(5H)-furan one)(5)And Isoramnetin(12)have a good inhibitory effect on Hela cells and Hep G2cells.Its inhibitory rates on Hela cells were 61.2%and 63.4%,and its inhibitory rates on Hep G2 cells were 62.9%and 53.6%,respectively.The isolated monomer compounds were screened forβ-lactamase inhibitory activity,when the sample concentration was at 1 mg/m L,the compound 3,5,7-trihydroxy-3′,4′-dimethoxyflavonoids(13)has a good inhibitory activity onβ-lactamase,with an inhibitory rate of 62.1%.
