Purpose:Using SP,M2 and TRPV1 as observation indicators,study the mechanism and targets of Qinbaiqingfei Concentrated Pills in reducing high airway response to cough after infection.Methods:1.The PIC guinea pig model was induced by smoke,capsaicin induced cough and peritoneal injection of a certain amount of LPS.2.The healthy guinea pigs were randomly divided into 4 groups.There are 10 animals in each group.Blank control group,model group(LPS 0.4mg/ml),positive control group,Qinbaiqingfei concentrated pill group.After the model was made,the PIC guinea pig were given intragastric administration for 10 days,and the number of coughs within 5 minutes was measured in the multi-purpose intelligent experimental animal gas fumigation method modeling device.After the experimental animals were anesthetized,the BALF and lung tissues were taken and the heart blood was taken.The content of SP,IL-6 and TNF-α were determined by ELISA.the expression of SP,M2 receptor and TRPV1 were determined by PCR and WB.Results:1.Comparison of cough frequency and time to first cough: Compared with the blank group,the number of coughs in the model group was significantly increased,and the time to first cough was earlier,the difference was statistically significant(P<0.05).Compared with the model group,the number of coughs in the Qinbai group and the control group were significantly reduced(P<0.05).There was no significant difference in the number of coughs between the control group and the Qinbai group(P>0.05).The first cough time of Qinbai group was significantly later than that of control group(P< 0.05).2.Comparison of the contents of IL-6,TNF-α,and SP in serum and BALF: Compared with the blank group,the contents of SP,TNF-α,and IL-6 in serum and BALF of the model group were significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,the contents of SP,TNF-α and IL-6 in serum and BALF of Qinbai group and Asime group were significantly reduced(P<0.05).Compared with the control group,the serum IL-6 and TNF-α levels in the Qinbai group were significantly reduced(P<0.05),but there was no significant difference in the serum SP levels between the two groups(P>0.05).Compared with the control group,the contents of TNF-α and SP in BALF of Qinbai group were reduced(P<0.05),but the contents of IL-6 in BALF were compared between the two groups(P>0.05)3.Comparison of SP in serum and SP and TRPV1 in BALF: Compared with the blank group,the serum SP contents in the model group and the SP and TRPV1 contents in BALF increased significantly(P<0.05).Compared with the model group,the serum SP content of Qinbai group and control group and the content of SP and TRPV1 in BALF were significantly reduced(P<0.05).Compared with the control group,the SP content in BALF in the Qinbai group was significantly reduced(P<0.05),but the serum SP content and the TRPV1 content in BALF were compared between the two groups(P>0.05)4.Comparison of M2 receptor and TRPV1 m RNA expression in lung tissue and BALF:Compared with the blank group,the TRPV1 and M2 receptor m RNA expression levels of the model group increased and decreased respectively(P<0.05).Compared with the model group,the expression levels of TRPV1 and M2 receptor m RNA in the BALF and lung tissues of the Qinbai group and the control group decreased and increased respectively(P<0.05).Compared with the control group,the expression level of M2 receptor m RNA in the Qinbai group increased(P<0.05)5.Comparison of the expression of M2 receptor and TRPV1 protein in lung tissue:Compared with the blank group,the expression of TRPV1 protein in the model group was increased and the expression of M2 receptor protein was decreased(P<0.05).Compared with the model group,the expression of TRPV1 protein in the Qinbai group and the control group was significantly decreased,and the expression of M2 receptor protein was significantly increased(P<0.05).Compared with the control group,the expression of M2 receptor protein in Qinbai group was increased(P<0.05),but the expression of TRPV1 protein in the two groups was compared(P>0.05).Conclusion:1.Qinbaiqingfei Concentrated Pills can reduce the content of inflammatory factors such as IL-6,TNF-α and SP,and multi-targets to reduce the inflammatory response in the PIC state,which can significantly reduce and delay the cough symptoms of PIC.2.Qinbaiqingfei concentrated pill can effectively treat PIC by reducing SP level,reducing TRPV1 content and expression,repairing and upregulating M2 receptor and other targets. |
