Background: The energy source of most malignant tumor cells is mainly aerobic glycolysis(Warburg effect),that is,even when the oxygen supply is sufficient,the glucose metabolism of the malignant tumor cells also tends to glycolysis.A number of studies have shown that C-Myc gene can regulate cell apoptosis and participate in the occurrence and development of a variety of malignant tumors.Studies have shown that long non-coding RNA nuclear enriched abundant transcript 1(lnc RNA NEAT1)is abnormally expressed in many cases,but the mechanism of its abnormal expression is not fully understood.Therefore,this experiment studied the regulatory relationship between C-Myc gene and long non-coding RNA NEAT1 in oral cancer cells,and the long non-coding RNA NEAT1 on the glycolysis level of Oral Squamous Cell Carcinoma(OSCC)Impact mechanism.Methods: Detect the expression level of the related long non-coding RNA in OSCC cells knocked down the C-Myc gene;then,detect the expression level of the long non-coding RNA NEAT1 up-regulated and knock-down C-Myc gene in OSCC cells;then,Up-regulate and knock down the expression of long non-coding RNA NEAT1 to detect the proliferation ability of OSCC cells;finally,up-regulate the expression of long non-coding RNA NEAT1,and detect the glycolytic metabolism of OSCC cells by glycolysis stress test.Results: Experimental results show that the expression of C-Myc gene and the long non-coding RNA NEAT1 is positively correlated,that is,the expression of the long non-coding RNA NEAT1 also shows an upward or downward trend when the C-Myc gene is upregulated or knocked down.In addition,by up-regulating or knocking down the expression of long non-coding RNA NEAT1,the proliferation ability of OSCC cells also showed the same trend;finally,up-regulating the expression of long non-coding RNA NEAT1 can increase the glycolysis level of OSCC cells.Conclusion: Therefore,this study predicts that C-Myc promotes the glycolysis level of OSCC cells by increasing the expression of lnc RNA NEAT1,thereby promoting the proliferation of OSCC cells and the development of oral cancer.
