Objective :To study the expression of miR-155 and NF-κB p65 in the mice model of ulcerative colitis(ulcerative colitis,UC)treated by bone marrow mononuclear cells(BM-MNCs)transplantation,and to explore the effect and mechanism of BM-MNCs transplantation on UCMethods :the 54 mice were randomly assigned to the normal control group,the UC model group and the BM-MNCs transplantation group,18 mice were used to extract BM-MNCs.The UC model group and the BM-MNCs transplantation group were modelled by the mice with the UC.And then the BM-MNCs transplantation were performed on mice with UC.The tissue damage situation on each mice group before and after the transplantation were observed using the Disease Activity Index and the ematoxylin and eosin(HE)stain.The miR-155 changes and the NF-κB p65 expression were tested using the rt-PCR technology and western blotting analysis respectively.Results :1.Compared to the normal control group whose DAI was always 0,UC model group DAI score was 2.72±0.102,Transplantation group score 1.28±0.102,the DAI of UC group and BM-MNCs transplantation group mice with the UC were remarkably increased.After the successful transplantation of the BM-MNCs,the DAI of the BM-MNCs transplantation group was significantly decreased in comparison with the UC model group.The differences have the statistics meaning(P<0.05).2.The mucosa was intact and the gland structure was normal arranged in the normal control group.Abundant neutrophil infiltrations,highly crypt cell damages,gland deformation and angiectasis below some mucosa were seen in the colon tissues of the UC model group.There were comparatively decreased neutrophil infiltrations,partly damages of the crypt cell in the transplantation group.the angiectasis or hyperaemia below the mucosa were unremarkable.Colonic mucosal tissue score,the normal control group is 0 points,UC model group score was3.62±0.31,BM-MNCs transplantation group score was 1.64± 0.35,transplantation group score was lower than those of UC model group,the two comparative differences are statistically significant(P<0.05).3.Regarding the expression of miR-155 in the mice colon tissues,UC model group was significantly increased compared to the normal control group,and the transplantation group was significantly decreased which tends to that of the normal control group in comparing with the UC model group.Three groups to compare the difference was statistically significant(P<0.05).4.Regarding the expression of NF-κB p65 in the mice colon tissues,UC model group was significantly increased compared to the normal control group,and the transplantation group was significantly decreased which tends to that of the normal control group in comparing with the UC model group.The UC group model group compared with the difference is statistically significant(P<0.05).Conclusions:1.DSS drinking can establish ulcerative colitis(UC)mice model;2.BM-MNCs transplanted into the mice UC model can migrate and colonize the damaged colonic tissue and have a repairing effect on the colonic mucosa of the lesion;3.BM-MNCs transplantation can inhibit the expression of miR-155 and NF-κB in UC colon tissue of mice.
