| Objective:Fucosyltransferase IV(FUT4)is a key enzyme responsible for the catalytic synthesis of fucosylated oligosaccharides Lewis X(Lex)and Lewis Y(Ley).Fut4 is highly expressed in many epithelial malignant tumors,and its expression characteristics and regulatory mechanisms are different in tumor cells.This study was to investigate the expression level of fucosyltransferase gene in children with acute lymphocytic leukemia(ALL)and its effect on prognosis.Method:(1)Bone marrow samples were collected from 110 children with acute lymphoblastic leukemia who were diagnosed in Jiangxi Children’s Hospital from January 1,2013 to December 31,2014,and analyz the clinical and laboratory characteristics。 The collected samples were agreed by the families of the children and the hospital ethics committee.(2)The total RNA was extracted from mononuclear cells by lymphocyte isolation medium.The concentration and purity of RNA were determined.The c DNA was reverse transcribed and stored at-80 ℃.(3)RT-q PCR was used to detect the expression level of FUT4 m RNA in 110 cases of all.Log rank test was used to analyze the 5-year survival rates and recurrence time of high / low expression group of FUT4.(4)Graphpad prism 5 software was used for data analysis and mapping.The test of homogeneity of variance and normality were carried out.T-test was used to compare univariate data between two groups;one-way ANOVA was used to compare multivariate data;nonparametric test was used to test uneven variance.The difference was statistically significant(P < 0.05).Results:(1)Among 110 ALL children,the median age was 4.33 years(range,0.42-15years).There were 64 males(58.18%).(2)There was no significant difference in the level of FUT4 between the two groups,but the number of leukocytes(63.02 ± 9.36)in the high expression group was significantly higher than that in the low expression group(31.00 ± 5.37),and the difference was statistically significant(P = 0.0416);(3)The expression of FUT4 m RNA in the newly treated all group was significantly higher than that in the control group((12.13 ± 2.36)vs(1.30 ± 0.33)).The expression of FUT4 m RNA decreased significantly after remission,and increased again after relapse;(4)In newly diagnosed ALL patients,the 5-year survival rate of high expression group of FUT4 was lower than that of low expression group(P = 0.0456),and the recurrence time was significantly shorter(P = 0.0323).Conclusion:The expression level of FUT4 was positively correlated with the progression of all.The recurrence rate of children with high expression of FUT4 was significantly higher,and the prognosis was poor.Thus,FUT4 gene can be used as one of the prognostic indicators of acute lymphoblastic leukemia.FUT4 is highly expressed in newly diagnosed and relapsed children with acute lymphoblastic leukemia,which may be a new biological indicator for judging residual leukemia. |
