| Objective:To investigate the expression of biological clock nuclear receptor REV-ERB αand lipid metabolism related protein SCD1 in head and neck squamous cell carcinoma(HNSCC)tissue and head and neck squamous epithelial tissue,and to analyze the relationship between the expression of REV-ERB α and SCD1 protein and the clinicopathological features of HNSCC patients,so as to explore whether REV-ERBα affects tumor progress by regulating the lipid metabolism of HNSCC.Methods:Immunohistochemistry was performed to analyze the REV-ERB α and SCD1 expressions in 50 HNSCC tissues and 40 head and neck squamous epithelium tissues.Kendall’s W test and Spearman correlation analysis were used to analyze the expression of REV-ERB α and SCD1 under different clinicopathological features and the correlation between them.Results:1.Immunohistochemical staining showed that the expression of REV-ERBα in HNSCC was significantly lower than that in head and neck squamous epithelium(P<0.001).The expression level of REV-ERBα was correlated with clinical stage(P=0.003)and lymph node metastasis(P<0.001),but not with sex,age,smoking and alcohol addiction,primary location of tumor and differentiation degree of tumor tissue(P >0.05).2.Immunohistochemical staining showed that the expression of SCD1 in HNSCC was significantly higher than that in head and neck squamous epithelium(P<0.001).The expression level of SCD1 was also correlated with clinical stage(P <0.001)and lymph node metastasis(P=0.007),but not with sex,age,smoking and alcohol addiction,primary location of tumor and differentiation degree of tumor tissue(P >0.05).3.Spearman correlation analysis showing the Negative correlation between REV-ERB α and SCD1 in HNSCC(rs=-0.434,P=0.002).Conclusions:1.The expression of REV-ERB α in HNSCC tissue was down-regulated,and its expression level decreased gradually with the increase of clinical stage of HNSCC,and it was related to lymph node metastasis or not,suggesting that REV-ERB α plays an important role in the occurrence,invasion and metastasis of HNSCC.It is promising to become a new target for HNSCC therapy.2.The expression of SCD1 was up-regulated in HNSCC,and its expression level gradually increased with the increase of clinical stage of HNSCC,and it was also related to lymph node metastasis,suggesting that the activity of lipid metabolism increased in HNSCC,and increased with the progression of tumor.3.There is a negative correlation between the abnormal expression of REV-ERBα protein and SCD1 protein,suggesting that REV-ERB α may affect the occurrence and development of HNSCC by regulating the lipid metabolism of HNSCC. |
