Research Of ABO Genes On ABO Subtype In Nanchang Population

ObjectivWe aimed to identify the ABO genes of ABO subtype through screening a large size of samples in Nanchang population and obtain the polymorphisms data of ABO alleles in Nanchang population,which will provide the data for ABO subgroup polymorphisms in Chinese Han population.Besides,The data can also guide ABO subtype blood transfusion in clinical to ensure the security of blood transfusion.Methods:We collected all samples of unpaid blood donors from Nanchang central blood station from 2017 to 2019.The donors should meet with the whole blood and component donor selection requirements.For the samples,which have discrepant results in forward /reverse ABO blood typing,serological tests were used to detect the ABO antigens and antibodies.Sequence-specific primer polymerase chain reaction(PCR-SSP)and sequence based typing(SBT)were used to detect the ABO genes.Results:81 681 samples were enrolled to this study.12 cases of ABO subtypes were found and the frequency was 1.47/10 000,including A subtype(5 cases: 2 cases in A blood group and 3 cases in AB blood group)and B subtype(7 cases: 5 cases in B blood group and 2 cases in AB blood group).The genotype of A subtypes were A201 and A205.The genotype of B subtypes were Bw03 and Bel03.Besides,we also found genotype O01/O01 in one B subtype samples.Conclusion:This study obtained the distribution data of ABO blood group gene polymorphisms in the population of Nanchang area,and provided guidance for blood transfusion of ABO subtypes Besides,it also proved that O genotype can cause weak B expression without other gene mutation.
