Background and Objective: Gastric cancer is the third leading cause of cancer death and the sixth most commonly diagnosed cancer in the world.In recent years,the diagnosis and treatment of gastric cancer has made great progress,but the overall survival rate of gastric cancer still needs to be improved.Studies have improved that circ RNAs is intently associated with a large amount of tumors,and they express differently in various diseases.But the mechanism remains unkown.This study aims to explore the expression,function and molecular mechanism of a new circ RNA(circ RNA_0074854)in gastric cancer.Patients and Methods: 102 pairs of gastric cancer tissues and adjacent nonmalignant tissues used in this study were all patients who received radical gastrectomy in the General Surgery Department of the Affiliated People’s Hospital of Jiangsu University from July 2016 to July 2019.We used RT-q PCR to evaluate the expression of circ RNA_0074854 in gastric cancer cell lines,gastric cancer tissues,and adjacent gastric nonmalignant tissues.The correlation between the expression level of circ RNA_0074854 and clinicopathological parameters was analyzed to verify its relationship with TNM stage,depth of invasion and lymph node metastasis.The function of circ RNA_0074854 in gastric cancer cells was analyzed by cell growth curve,colony formation,Transwell method and flow cytometry in vitro.Western blot was used to analyze the effect of circ RNA_0074854 on the expression of Epithelial-mesenchymal transition(EMT)related proteins.Results: The results of RT-q PCR showed that circ RNA_0074854 was downregulated in gastric cancer tissues and cell lines.Clinical correlation analysis showed that circular RNA_0074854 was distinctly negatively connected with invasion depth,TNM stage and lymph node metastasis in gastric cancer tissues.In function,circ RNA_0074854 inhibited cell proliferation,migration and invasion,induce cell apoptosis,and block gastric cancer cells in G1 phase.The expression of vimentin and N-cadherin proteins was inhibited.Conclusions: The results of RT-q PCR showed that the circ RNA_0074854 was in a low expression level in gastric cancer tissues.Clinicopathological correlation analysis showed that the expression level of circ RNA_0074854 in gastric cancer tissues was distinctly negatively connected with invasion depth,TNM stage and lymph node metastasis.Cell function experiments indicated that interference with circ RNA_0074854 promoted proliferation,migration and invasion of gastric cancer cells,block cell cycle progression,and inhibit cell apoptosis.Similarly,overexpression of circ RNA_0074854 can inhibit cell proliferation,migration and invasion,inhibit cell cycle progression and promote cell apoptosis.Western blot results showed that EMT was inhibited by inhibiting the expression of vimentin and N-cadherin proteins.Therefore,circ RNA_0074854 is expected to be a new molecular marker for the diagnosis and treatment of gastric cancer.Therefore,circ RNA_0074854 is expected to be a novel molecular marker for both the diagnosis and treatment of gastric cancer. |
