| Objective:Lung cancer is the most common with the highest mortality rate in China.Only18% of patients diagnosed with lung cancer can survive for 5 years or more.The diagnosis of lung cancer mainly relies on imaging and pathological examination,among which invasive pathological biopsy is the gold standard for the diagnosis and classification of lung cancer.The early symptoms of lung cancer are not obvious,and laboratory molecular markers are lack of specificity.CircularRNA(circRNA)discovered in recent years is a novel Competing endogenousRNA(ceRNA).Because of its stability,conservatism,universality,tissue and disease specificity,circRNA is expected to become a biomarker for cancer diagnosis and prognosis evaluation.In recent years,there have been many reports on the role of circRNAs in the diagnosis and prognosis of cancer,such as gastric cancer,breast cancer and glioma.In this study,circRNAs differentially expressed in the tissues of lung adenocarcinoma patients were screened and verified,and the value of hsa_circ_0001320 as a molecular biomarker for early diagnosis of lung adenocarcinoma was preliminarily discussed.Methods:1.Screening of differential expression profiles circRNA lung adenocarcinoma :Hsa_circ_0001320 was selected as the study object by high-throughput sequencing in different lung tissues.CircRNA RNase and agarose gel electrophoresis were used to identify the structure of hsa_circ_0001320.2.Verifying the expression levels: The expression levels of hsa_circ_0001320 in lung adenocarcinoma tissues,adjacent tissues and lung adenocarcinoma cellsperipheral blood serum were detected by qRT-PCR.Statistical methods were used to analyze the expression of hsa_circ_0001320 in lung adenocarcinoma tissues and cells.3.The expression levels of hsa_circ_0001320 in peripheral blood serum were detected by qRT-PCR.Statistical methods were used to analyze the relationship between hsa_circ_0001320 and clinical characteristics.A receiver operating characteristic(ROC)curve analysis was used to analyze the diagnostic accuracy of hsa_circ_0001320.Results:1.The expression of hsa_circ_0001320 is downregulated in lung adenocarcinoma tissue and human lung adenodenocarcinoma cells.2.The expression of hsa_circ_0001320 is downregulated in peripheral blood serum.3.The expression of hsa_circ_0001320 in peripheral blood is negatively correlated with lymph nodes and distant metastasis.4.The area under the ROC curve was 0.752.Conclusion: The expression of hsa_circ_0001320 is downregulated in lung adenocarcinoma,and it could be used as a potential biomarker for the early detection of lung adenocarcinoma. |
