TRPC5 is a member of the transient receptor potential(TRP)non-selective cation channel family.It plays an important role in many physiological processes.It is related to cardiovascular disease,central nervous system disease and kidney disease.Previously,our group analyzed the Cryo-EM structure of TRPC5 in PMAL-C8,but it’s structure in the nanodiscs has not been solved yet.Because the use of traditional detergents to display intact membrane proteins may result in the loss of part of the information about the interaction of proteins and lipids,this study uses cryo-electron microscopy technology and phospholipid bilayer nanodisc reconstitution technology to show the structure of TRPC5 in natural lipid bilayers.At the same time,TRPC5 is assembled into nanodiscs,which can be used as an antigen for alpaca immunity to obtain nanobodies for the treatment of related diseases.
