| In tissue engineering,silicate-based bioactive glass(BG)can promote the repair of a variety of tissues and has gained widespread clinical application.In other fields,such as botany and nutrition,Si ions are considered to promote the growth of plant and animal bones.Si and silicate-based materials have a promoting effect on a variety of cells,also activate many biological processes.While the mechanism by which it exerts its biological functions is still unclear.Some studies attributed the biological activity of Si and silicate-based materials to the activation of specific biological signals by Si ions,but that was not enough to explain the biological effects of Si ions on multiple cells and multiple signaling pathways.Here we explain the effects of Si and silicate-based bioactive glass from the perspective of Si ion affecting cell membrane fluidity,which may explain the effect of Si and bioactive glass on various cells and the activation of multiple signaling pathways.First,through the fluorescence bleaching recovery technology and the overall cell membrane polarizability measurement experiments,we had proved that Si ions effectively affect the cell membrane fluidity in a concentration-dependent manner.Further study the mechanism of Si affecting cell membrane fluidity.Remove or add Si-containing medium for a short period of time,the cell membrane fluidity changes rapidly and reversibly,indicating that Si ions can directly regulate cell membrane fluidity by directly affecting the cell membrane.Also,Si ions do not affect the lipid structure of the liposome,and only enhance the movement of lipid molecules on the liposome,indicating that Si enhances the fluidity of the cell membrane through a physical way.A large number of papers have reported the enhancement of HUVEC cell migration ability and the promotion of osteogenic differentiation of HBMSC cells by Si-containing bioactive glass ion extract.By specifically inhibiting cell membrane fluidity with cholesterol succinate monoester(CHS),the biological activity of Si-containing bioactive glass ion extract is completely inhibited.Migration and differentiation of cells co-treated with CHS and Si were inhibited to the same level as those treated with CHS alone,which shown that the Si-containing bioactive glass ion extract mainly exerts its biological activity through the cell membrane.Based on the theory that Si ions affect cell membrane fluidity,we forecast that Sicontaining BG ion extracts promote apoptosis of injured cells,especially the early apoptosis of cells marked by the lipid membrane phosphatidylserine eversion.The gene expression and protein activity chip detection of HBMSC cells treated with Si ions also supports that Si ions pass through the cell membrane to widely affect cell signal transduction.Monte Carlo simulation of cell membrane fluidity and signal transduction,further explaining the effect of cell membrane fluidity on cell signal strength and cell responseOverall,this study has proved that Si ions exert their bioactivities on cells by directly enhancing cell membrane fluidity and subsequently affect,which provided an explanation for the general bioactivities of silicate material. |
