| Objective: To screen high risk genes in gastric cancer using public database;to determine the transcriptional and translational expression levels of this gene in gastric cancer cell lines and tissues;to investigate the effect of this gene on clinical prognosis of patients;to describe the molecular mechanism of its coding protein in the microenvironment of gastric cancer,providing clues and ideas for the early clinical diagnosis and targeted intervention of gastric cancer.Methods: Paired gastric cancer tissue samples were selected from TCGA database,and the high-expression gene CLDN6 was screened out by bioinformatics analysis.CLDN6 expression in different cell lines was detected in gastric cancer cell lines,and CLDN6 expression in cancer tissues and paracancer tissues was verified in tissue samples from patients in Renji hospital and public GEO data.The effects of CLDN6 on the proliferation and invasion of gastric cancer cells were investigated by in vitro cell function experiment and in vivo animal tumor formation experiment.GSEA analysis of GEO public database was used to find the signaling pathways that might be affected by CLDN6,and western blot,PCR,immunoprecipitation,cellular immunofluorescence Ch IP and other experiments were conducted to verify the changes of key proteins and genes.Conclusion: CLDN6 is highly expressed in gastric cancer tissues compared with adjacent tissues,and high expression of CLDN6 indicates poor prognosis of patients.CLDN6 can promote the proliferation and invasion of tumor cells through YAP1-snail1 axis. |
