| Objective:To research the effects of isoliquiritigenin unite temozolomide on the vasculogenic mimicry,cell proliferation ability and migration distance of SHG44 glioma cells in hypoxic environment and research it’s possible mechanism.This experimental research is innovative and forward-looking to a certain extent.Methods:Under the hypoxic environment simulated by cobalt dichloride,SHG44 was treated with blank control,isoliquiritin(160umol/L),temozolomide(200umol/L),isoliquiritin combined with temozolomide(160umol/L+200umol/L),respectively Glioma cells 24 h,use threedimensional culture method to detect the ability of SHG44 glioma cells to generate angiogenesis mimicry;use CCK-8 test kit to measure the proliferation ability of SHG44glioma cells;use cell scratch test to measure the migration distance of SHG44 glioma cells.SHG44 glioma cells was determined by western blotting technology to determine the proteins related to vasculogenic mimicry: epithelial cell kinase A2(rythropoietin-producing hepatocellular,EphA2),matrix metallopeptidase-2(MMP-2)and vascular endothelial growth factor(Vascular Endothelial Growth Factor,VEGF)expression levels.Results:1.The first part experiment:Compared with the blank control group,isoliquiritin and temozolomide single agent group in hypoxic environment,the number and length of angiogenesis mimicry of SHG44 glioma cells in the isoliquiritin + temozolomide group decreased more significantly,and the cell proliferation ability was reduced It is more significant,the migration distance of cells is also reduced more significantly,and the difference is statistically significant(P<0.05).2.The 2.The2.second part experiment:Obtained by western blotting experiment that in hypoxic environment,the expression content of VEGF,EphA2 and MMP-2 protein of glioma cells in the blank control part,isoliquiritin group and temozolomide group did not change significantly,and the difference was not statistically significant(P>0.05).The expression levels of MMP-2,VEGF,and EphA2protein in temozolomide group did not change significantly in the hypoxia environment,the difference was not statistically significant(P>0.05).In the isoliquiritigen+ temozolomide group,the expression levels of MMP-2,VEGF,and EphA2 protein decreased significantly,and the difference was statistically significant(P<0.05).Conclusions:1.The first part experimentIsoliquiritigen combined with temozolomide can inhibit the proliferation and migration of SHG44 glioma cell VM and cells inhypoxia environment.2.The second part experimentUnder hypoxia,isoliquiritigenin combined with temozolomide can inhibit the angiogenesis mimicry and cell proliferation and migration of SHG44 glioma cells.The reason may be that it reduces the expression of MMP-2,VEGF and EphA2 protein. |
