Objective:To investigate the expression frequency and immunomodulatory effects of regulating B cells and IL-10 in GBS patients and healthy controls.At the same time,observe their relationship with the severity of disease in GBS patients.It provides certain theoretical feasibility for regulating B cell targeted therapy of GBS.Methods:From December 2019 to February 2021,13 GBS patients and 13 healthy volunteers who were hospitalized in the First Affiliated Hospital of Bengbu Medical College,Anhui Province were selected,and the GBS patients were scored with Hughes function.Two types of regulatory B cells were detected by flow cytometry,and the counts of CD19+CD24hiCD38hiBreg cells and CD19+CD24hiCD27+Breg cells were obtained.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of IL-10 in the peripheral blood of both.SPSS software was used to detect the difference between GBS patient group and healthy control group.Pearson grade correlation coefficient was used to analyze the relationship between regulatory B cells and IL-10 and Hughes function score.Results:The peripheral CD19+CD24hiCD38hiBreg cell count in GBS group was(3.57±1.57)%,the count in healthy control group was(7.56±1.62)%,and the peripheral blood CD19+CD24hiCD27+Breg cell count in GBS group was(15.68±8.21)%,that in healthy contiol group was(21.27±2.74)%.Compared with healthy control group,regulatory B cells in GBS group decreased signifucantly(all P<0.05).The level of IL-10 in peripheral blood of GBS group was(155.46±146.09)pg/ml,and the count in the healthy control group was(5.20±2.60)pg/ml.The GBS group was dozens of times higher than the healthy control group(P<0.05).In the GBS group,peripheral blood CD19+CD24hiCD38hiBreg cells,CD19+CD24hiCD27+Breg cells and IL-10 were not correlated with the severity of disease in GBS patients(all P>0.05).Conclusion:1.CD19+CD24hiCD38hiBreg cells,CD19+CD24hiCD27+Breg cells and IL-10 may play a key role in the pathogenesis of GBS.2.CD19+CD24hiCD38hiBreg cells,CD19+CD24hiCD27+Breg cells may down-regulate the immune response of GBS,and it may be mediated by IL-10.
