Objective:This topic using Morris water maze test navigation experimental rats escape latency and space exploration experiment target quadrant residence time and through the original table number,transmission electron microscopy(sem)observation synaptic ultrastructure of hippocampus CA1 area neurons,Western blot detection in the rat hippocampus PSD-95 with Me CP2 protein expression,to explore the electricity to improve the mechanism of action of sleep-deprived rats cognitive.Methods:A total of 36 male SPF SD rats were randomized into 4 groups.Blank control group: When the other 3 groups were modeled,the rats in this group were raised in a large platform water environment.When the rats in the electroacupuncture group and the sham electroacupuncture group were acupunctured,the rats in this group were grabbed by the same method.Model group: Improved multi-platform water environment method was used to prepare the REM sleep deprivation model of rats.When the rats in the electroacupuncture group and the sham electroacupuncture group were acupunctured without acupuncture intervention,the rats in this group were grabbed by the same method.Electroacupuncture group: Improved multi-platform water environment method was used to prepare the REM sleep deprivation model of rats,and electroacupuncture intervention was given every day during the modeling process.The sham electroacupuncture group: improved multi-platform water environment method was used to prepare the REM sleep deprivation model of rats,and the sham electroacupuncture was given daily intervention during the modeling process.On the first day of the sleep deprivation experiment,the electroacupuncture treatment was performed at 9:00 am every day at the acupuncture group’s "Baihui" and "Dazhui" acupoints.The sham electroacupuncture group took the non-acupoint area of 3mm next to the "Baihui" and "Dazhui" acupoints as the acupuncture point.After connecting the electroacupuncture instrument,no power supply was turned on.Electroacupuncture group and sham acupuncture group each time for 15 min,once a day,for 5 consecutive days.Morris water maze was used to detect the escape incubation period and the target quadrant residence time of the positioning navigation experiment and the number of times of crossing the original platform.Transmission electron microscopy was used to observe the synaptic ultrastructure in the CA1 region of the hippocampus,and Western blot was used to detect the expressions of PSD-95 and Me CP2 proteins in the hippocampus of the rats.Results:1.General status observation: compared with the blank control group,the rats in the model group showed listlessness,less activity,messy and dull body hair,increased aggression,and significantly decreased body weight(P<0.05).There was no significant difference in mental state between the model group and the sham group,and there was no statistically significant difference in body weight(P>0.05).Compared with the model group,the rats in the electroacupuncture group had better mental state,more flexible response,less aggression and weight gain(P<0.05).2.Behavioral results: Compared with the results of positioning navigation test of rats in each group,the avoidance latency of rats in the model group was significantly increased(P<0.05).Compared with the model group,the evasive latency of electroacupuncture group was significantly shortened(P<0.05).There was no significant difference between the sham electroacupuncture group and the model group(P>0.05).Compared with the blank control group,the time of staying in the target quadrant of the model group was significantly shortened(P<0.05),and the number of crossing the original platform was significantly reduced(P<0.05).There was no significant difference between the sham group and the model group in terms of the time to stay in the target quadrant and the number of times to cross the original platform(P>0.05).Compared with the model group,the rats in the electroacupuncture group stayed in the target quadrant for a longer time(P<0.05),and the number of crossing the original platform increased significantly(P<0.05).3.Transmission electron microscopy results: In the blank control group,the number of synapses was relatively rich,most of the synapses were not significantly edema,the membrane was intact,the synaptic structure was intact and clear,the synaptic gap width was moderate,the presynaptic membrane and the posterior synaptic membrane were clear,the post-synaptic dense area was thickened and continuous,and the synaptic vesicles were abundant.Compared with the blank control group,the model group showed a decrease in the number of synapses,mild edema of the synaptic corpuscles,complete fusion of half of the synaptic structures,unclear presynaptic membrane and posterior membrane,narrowing of the synaptic gap,uneven thickness of the post-synaptic dense area,and reduced number of synaptic vesicles.There was no significant difference between the sham group and the model group.Compared with the model group,the electroacupuncture group showed an increase in the number of synapses,more complete synaptic structure,clear presynaptic membrane and posterior membrane,moderate synaptic gap width,moderate post-synaptic density area,longer synaptic interface,and more synaptic vesicles.4.Western blot results: the overall trend of the relative expressions of PSD-95 and Me CP2 was consistent.Compared with the blank control group,the relative expressions of PSD-95 and Me CP2 in the hippocampal tissues of the model group were significantly decreased(P<0.05).Compared with the model group,the relative expression levels of PSD-95 and Me CP2 were higher in the hippocampal tissues of the electroacupuncture group(P<0.05).There was no significant difference in the relative expression levels of PSD-95 and Me CP2 in hippocampal tissues between the sham group and the model group(P>0.05).Conclusion:1.Electroacupuncture treatment can significantly improve the learning and memory ability of sleep-deprived rats.2.Electroacupuncture can improve the synaptic ultrastructure in the hippocampus CAl region of SD model rats and maintain the integrity of the synaptic structure.3.Electroacupuncture may improve the learning and memory of SD model rats by up-regulating the expression of PSD-95 and Me CP2 proteins,improving the synaptic ultrastructure of hippocampal neurons,and promoting the enhancement of synaptic plasticity.
