Long non-coding RNA(lnc RNA)plays an important regulatory role in the life of human cells.Numerous studies have shown that human diseases are closely related to the regulatory dysfunction of lnc RNA.While the study of lnc RNA started late,the regulatory function of lnc RNA and its association with diseases are not very clear.With the deepening of studies on lnc RNA,there are more and more reports about the regulatory functions of lnc RNA and the association between lnc RNA and human diseases,which provides great convenience for the research of regulatory functions of lnc RNA.This paper proposed the bioinformatics methods of lnc RNA enrichment analysis in the form of set for the first time,based on collecting and sorting out a large number of relevant literatures about lnc RNA,aiming at the problem of low functional efficiency of lnc RNA analyzed by experimental methods,which greatly improved the efficiency of analyzing lnc RNA regulatory function.In addition,in order to more comprehensively analyze the regulatory relationship of user-input lnc RNA sets in different diseases,a disease similarity calculation method based on the correlation strength of lnc RNA-human diseases was proposed,so as to comprehensively understand the disease regulatory function of user-input lnc RNA sets.The main contents of this paper are as follows:(1)This paper firstly proposed a set-based lnc RNA enrichment analysis system.It collected and sorted out the data of lnc RNA regulation function and its association with diseases by manual collection and download of public database data,and classified lnc RNA by function and disease set.On the basis of the lnc RNA set data,the hypergeometric distribution method was used to carry out the enrichment analysis of the functional and disease association relationship of the lnc RNA set data input by users,and the analysis results were represented by P value.In order to improve the accuracy of the result analysis,FDR verification and Bonferroni correction were carried out on the results.(2)This paper firstly proposed a method for calculating disease similarity based on lnc RNA-human disease correlation strength.In order to further analyze the regulatory effect of lnc RNA on diseases,the regulatory relationship was subdivided into up-regulation and down-regulation relationships,and the lnc RNA-human disease correlation strength matrix was established.When users input the lnc RNA set,the lnc RNA-human disease correlation strength vector of the set is constructed in first,and then the similarity between the vector and the different disease correlation strength vectors in the lnc RNA-human disease correlation strength matrix is calculated by using the cosine similarity formula,which provides help for the comprehensive and in-depth analysis of the disease regulation function of lnc RNA.In order to facilitate the use of the aforesaid methods,LSEA(lnc RNA Set Enrichment Analysis)of web server version is designed and implemented in this paper.The system has a friendly interface and perfect functions.The lnc RNA set input by the user not only realizes the function enrichment analysis and related disease enrichment analysis,but also realizes the similarity analysis function of lnc RNA set regulating human diseases,and visualizes the analysis results.In order to adapt to the growing number of lnc RNAs,the system also added the query function to facilitate users to quickly understand the regulatory functions of interesting lnc RNAs.
