| Purpose : Observing the effect of Xiezhuoqingjie decoction on the general condition,Disease Activity Index(DAI)score,histopathological changes of colon,content of serum Interleukin-22(IL-22),expression of IL-22 and content of MDA in colon tissue of ulcerative colitis(UC)rats,to explore the possible mechanism of Xiezhuoqingjie decoction in treating UC.Method:Sixty Wistar rats were randomly divided into six groups:Blank Group,Model Group,Western Medicine Control Group,High Dose Group,Medium Dose Group,Low Dose Group.In addition to the Blank Group,the other five groups were eatablished UC models by 2,4,6-trinitrobenzene sulfonic acid(TNBS)/ethanol method.After modeling,the Model Group was given normal saline,the other four groups were given the corresponding concentration of drug suspension gavage for two weeks.In the meantime,the general condition of rats was observed and the DAI score was performed.The rats were killed after the last administration of the drug,the blood of femoral artery and colon tissue were taken,the pathological morphology of colon was observed and scored.The content and expression of IL-22 was detected by ELISA,RT-PCR and Western Blot methods.The content of MDA in colon was detected by thiobarbituric acid.Results:1.General condition and DAI scoreRats in Blank Group were in good spirit,with shiny fur,normal water intake,normal weight gain and normal urination and defecation.The other five groups’ rats were depressed,the fur was dull,the water intake was reduced,the weight loss,the stool was watery with pus and blood.After the drug treatment,the general condition of each group was improved,but worse than the Blank Group.DAI scores show: All the drug treatment groups were lower than the Model Group(P<0.05);The High Dose Group was lower than the Medium Dose Group,Low Dose Group and the Control Group(P<0.05);The Medium Dose Group and the Control Group had no significant difference(P>0.05),and both groups were lower than the Low Dose Group(P<0.05).2.Histopathologic changes of colonThe colon mucosa of the Blank Group was normal;In the Model Group,there were extensive ulceration,obvious hyperemia and edema.The mucosal ulceration was improved in all drug treatment groups,with the most significant improvement in High Dose Group,the least improvement in Low Dose Group,and the mucosa of Medium Dose Group was similar to that of Control Group.3.Target detectionLevel and expression of IL-22,content of MDA:The Blank Group was lower than other groups;The treatment groups were lower than the Model Group(P<0.05);The High Dose Group was lower than the other drug treatment groups(P<0.05);There was no significant difference between the Medium Dose Group and the Control Group(P>0.05),and both groups were lower than the Low Dose Group(P<0.05).Conclusion:Xiezhuoqingjie decoction can obviously improve the general condition of UC rats and promote the mucosal repair.The dose of Xiezhuoqingjie decoction has correlation with the curative effect.The Therapeutic mechanism may be related to decreasing the content and expression of IL-22 and the content of MDA,thus promoting the healing of intestinal mucosa and reducing inflammation. |
