| Objective: To study the mechanism of METTL5 in tumors,predict the miRNAs involved in the regulation of liver cancer expression in METTL5,so as to find research targets for liver cancer treatment.Methods: This study through the analysis of the generic carcinoma combined multiple database to reveal the mechanism of action of METTL5,through to the METTL5 gene expression in queue and a variety of Tumor cell lines,survival prognosis,immune cell infiltration analysis,Tumor mutational burden(TMB)analysis,Microsatellite instability(MSI),immune associated checkpoint gene expression analysis,Mis-match Repair(MMR)gene expression and DNA methylation gene expression related to correlation analysis,To comprehensively study the gene expression distribution pattern and gene function of METTL5 in tumor.Then on N6-methyl adenine(N6-methyladenosine m6A)methylation regulating gene expression in liver cancer and prognosis were analyzed,and concluded that its expression in liver cancer and prognosis,further through TCGA database,access to liver cancer Micro RNAs(miRNAs)expression profile chip,analyze differentially expressed,miRNAs have differences,and then to forecast analysis can combine METTL5 Micro RNAs,miRNAs will predict the miRNAs and differences between intersections,Target miRNAs of METTL5 in liver cancer were obtained.Next,differential miRNAs target genes were predicted and screened to obtain co-regulatory genes of METTL5 targeted by differential miRNAs.Finally,GO/KEGG prediction and enrichment analysis of target genes were performed to explore the functions and related signaling pathways of miRNAs co-expression regulation genes involved in the regulation of METTL5 expression.Results: In this study,the mechanism of action of METTL5 was revealed through the combination of multi-database Pancancer analysis,and it was found that METTL5 was abnormal in a variety of tumors,affected the survival and prognosis of patients,and was involved in immune cell infiltration,TMB,MSI,immune checkpoint genes,DNA repair and DNA methylation in a variety of tumors.Then,the expression and prognosis of M6 A methylation regulatory genes in HCC were analyzed.It was found that most of the genes involved in M6 A methylation regulatory genes were not significantly expressed,but METTL5 was upregulated in HCC,and these regulatory genes affected the prognosis of HCC patients.Further differential expression analysis was performed to obtain differential miRNAs,and the prediction analysis of target genes of differential miRNAs was performed to obtain the interaction relationship between miRNAs and METTL5,and hsa-miR-511-5p was screened to be an effector molecule involved in the regulation of METTL5 expression in liver cancer.Then,the GO/KEGG prediction and enrichment analysis of hsa-miR-511-5p target genes were performed to identify the key molecular functions,cell components and biological processes involved in hsa-miR-511-5p.It was found that the miRNA co-expression regulation genes involved in the regulation of METTL5 expression were related to multiple pathways and functions such as growth factors,mechanical stress,and cell metabolism.Conclusions:1.METTL5 is generally highly expressed in pan-carcinomas,and is detrimental to the prognosis and survival of patients.it is widely involved in immune cell infiltration,TMB,MSI,immune checkpoint genes,DNA repair and DNA methylation in a variety of tumors.2.Hsa-miR-511-5p is an effector molecule involved in the regulation of METTL5 expression in liver cancer. |
