Objective: To study the effect of abnormal expression of Integrin-linked kinase(ILK)in esophageal squamous cell carcinoma cells on the sensitivity of cisplatin chemotherapy at the cellular level,providing a theoretical basis and new strategies for precise treatment of esophageal malignant tumors.Method:1.Online analysis of the expression of ILK in esophageal squamous cell carcinoma and normal esophageal tissues in the Cancer Genome Atlas(TCGA)database,and divide the abnormal expression of ILK in esophageal squamous cell carcinoma into high expression and low expression In the expression group,compare the difference in survival time between the two groups.2.The established esophageal squamous cell carcinoma cell lines with over-expression and interference with successful transfection were screened out,and the effect of abnormal expression of ILK in esophageal squamous cell carcinoma cells on cisplatin chemotherapy sensitivity was evaluated by the CCK8 experimental method.Results:1.The ILK expression in cancer tissues of 184 cases of esophageal squamous cell carcinoma in the TCGA database was significantly higher than that in 11 cases of normal esophageal tissues(p<0.05),and the difference was statistically significant;In patients with esophageal squamous cell carcinoma,the survival time of patients with high ILK expression was shortened(p>0.05),but the difference was not statistically significant.2.Under the condition of different cisplatin concentration,the inhibition rate of ILK overexpression group was lower than that of control group.The IC50 value of the ILK overexpression group(16.78±0.41)was significantly higher than the IC50 value of the control group(8.33±0.37)(p<0.05);3.Under the condition of different cisplatin concentration,the inhibition rate of ILK interference expression group increased compared with the control group high.The IC50 value of ILK interference expression group(3.72±0.52)was significantly lower than that of the control group(8.74±0.46)(p<0.05).Conclusion:ILK is highly expressed in esophageal squamous cell carcinoma;Cytological experiments have confirmed that overexpression of ILK can reduce the sensitivity of esophageal squamous cell carcinoma to cisplatin chemotherapeutics;while interference with ILK expression can increase the sensitivity of esophageal squamous cell carcinoma to cisplatin chemotherapeutics.
