Objective:To identify the specific biomarkers and explore the signal pathways involved in acute myocardial infarction by analyzing the expression profile of non-coding RNAs in peripheral blood of patients with AMI via RNA sequencing technology.Methods:The whole blood of anterior cubital vein from 3 patients with ST segment elevation myocardial infarction and 3 non-coronary heart disease controls were analyzed with RNA sequencing technology to explore the expression characteristics of lncRNA,circRNA and mRNA.The GO,KEGG pathway enrichment analysis,disease association analysis and DO disease enrichment analysis of mRNA were carried out through GO database,KEGG database and VarElect databases,and then the protein-protein interaction network was constructed from STRING database to screen key genes.Starbase database was used to predict the targeted miRNAs of key genes and differential lncRNA and circRNA.combined with cytoscape software to display the CeRNA network map of lncRNA-miRNA-mRNA and circRNA-miRNA-mRNA was constructed by cutoscape software to explore the pathogenesis and potential signal pathways involved in AMI from the perspective of lncRNA/circRNA-miRNA-mRNA interaction.Results:This study identified 461 mRNAs with 241 up-regulated and 220 down-regulated,516 lncRNA with 236 up-regulated and 280 down-regulated,11 circRNA with 2 up-regulated and 9 down-regulated.Differentially expressed mRNAs were analyzed with GO enrichment,KEGG pathway enrichment,disease association enrichment and PPI network were constructed to finally acreen out 3 key genes closely related to AMI:CCL5,CCR5,CX3CR1.Seven miRNAs were found to have a coexpression relationship with CCL5 and CCR5 with upstream miRNA prediction.Seven interaction miRNAs were shared by eleven differential lncRNA and the two genes through the interaction miRNA prediction of differential lncRNA.Similarly,the interaction miRNA prediction of 11 differential circRNA showed that 4 interaction miRNAs were shared by 4 differential circRNA and the two genes.Finally,ceRNA networks of lncRNA-miRNA-mRNA and circRNA-miRNA-mRNA models were constructed to analyze the pathogenesis of acute myocardial infarction.Conclusions:1.A large number of immune-related biological processes and cardiovascular disease-related metabolic pathways were obtained by functions and pathways enrichment of mRNA,indicating that immune and cardiovascular-related genes may play an important role in the occurrence and development of AMI.2.The chemotaxis,activation and inhibition of inflammatory cells by CCL5 and CCR5 are closely related to the occurrence and development of acute myocardial infarction.It is speculated that CCL5 and CCR5 can be used as target genes and new therapeutic targets for serum markers of AMI to judge the degree of cardiomyocyte injury or the prognosis of myocardial infarction.3.The expression of 11 lncRNA,4 circRNA and 2 key genes CCL5 and CCR5 were down-regulated in the constructed lncRNA/circRNA-miRNA-mRNA ceRNA network,and 4 common miRNAs were predicted by the two networks.It is suggested that 11 lncRNA and 4 circRNA can be used as ceRNA to regulate the expression of CCL5 and CCR5 by competitive binding of miRNAs.These non-coding RNAs may be used as potential biomarkers and therapeutic targets for acute myocardial infarction.
