| Objectives:To explore the effects of gastrodin on the M2 polarization and MAPK pathway of lipopolysaccharide(LPS)activated BV-2 cells,and whether gastrodin can regulate the polarization of M2 BV-2 cells through the p38MAPK pathway.Methods:BV-2 microglia cell lines were cultured in vitro,and BV-2 microglia cells were activated by LPS and divided into three groups:control group,LPS activated group,and LPS+gastrodin pretreated group.The expression of markers(IL-10,Argl,YM1/2 and CD206)and MAPK-related proteins(p-p38,P38,p-JNK,JNK,p-ERK1/2 and ERK1/2)in M2 type BV-2 cells were detected by Western Blot and fluorescence double-label staining.After pretreatment with a p38MAPK pathway inhibitor(SB203580),The P-p38 protein and M2-type BV-2 cell marker IL-10 and YM1/2 in the control group,SB203580 group,LPS activated group,LPS+SB203580 group,LPS+ Gastrodin pretreatment group and LPS+ SB203580+ Gastrodin pretreatment group were detected by Western Blot.Results:(1)Western Blot and fluorescence staining results showed that the protein expressions of IL-10,Argl,YM1/2 and CD206 were lower in the control group,while the expression levels in the LPS group were significantly higher than those in the control group,and the results were statistically significant(p<0.05);The expression of IL-10,Argl,YM1/2 and CD206 were significantly increased after gastrodin intervention.Compared with the LPS group,there was a significant difference(p<0.05).(2)Western Blot and fluorescence staining showed that the expression levels of p-P38,p-ERK1/2 and p-JNK in LPS group were significantly higher than those in the control group(p<0.05);The expression of p-P38,p-ERK1/2 and p-JNK decreased significantly after gastrodin intervention(p<0.05).P38,JNK and ERK1/2 had no significant differences among groups(p>0.05).(3)Western Blot results showed that the expression of p-P3 8 protein was decreased and the expression of YM1/2 and IL-10 were significantly increased after the treatment of gastrodin and P38 inhibitor(SB203580),which was different from that of gastrodin preintervention group(p<0.05).Conclusions:(1)Gastrodin intervention significantly promoted the expression of LPS-activated BV-2 cells M2-type microglia markers IL-10,Argl,YM1/2 and CD206.(2)Gastrodin intervention significantly reduced the expression of p-P38,p-ERK1/2 and p-JNK in LPS-activated BV-2 cells.(3)Gastrodin may regulate the M2-type polarization of BV-2 cells activated by LPS through the P38MAPK signaling pathway. |
