| Objective:To investigate the expression ofβ-catenin in Wnt/β-catenin signaling pathway in pancreatic ductal adenocarcinoma(PDAC)tissues,and to investigate the relationship betweenβ-catenin expression and clinicopathological characteristics and prognosis of patients.Methods:Primary pancreatic cancer cell lines and normal human pancreatic duct epithelial cell lines(HPDE6-C7)were cultured,and total RNA was extracted.The expression level ofβ-catenin m RNA in primary pancreatic cancer cells and HPDE6-C7 cell lines was detected by real-time quantitative PCR.Forty-five pairs of cancer tissue and paracancerous tissue samples from patients with pathologically confirmed PDAC in our hospital from June 2012 to December 2013 were collected.The expression level ofβ-catenin in cancer tissue and paracancerous tissue was analyzed by immunohistochemical method,and the relationship between the expression level ofβ-catenin in cancer tissue and clinicopathological characteristics and prognosis was analyzed.Results:Compared with the control HPDE6-C7 cell line,the m RNA relative expression ofβ-catenin in primary pancreatic cancer cell line was significantly increased,the difference was 4.32(P<0.05),with statistical significance;The expression level ofβ-catenin in cancer tissue of PDAC patients was higher than that of paracancer group(χ~2=7.50,P=0.005),and the difference was statistically significant.The expression level ofβ-catenin was positively correlated with tumor size and TNM stage in PDAC patients(P=0.026,0.036).Kaplan-Meier survival analysis showed that the overall survival rate in the group with highβ-catenin expression was lower than that in the group with lowβ-catenin expression,and the difference was statistically significant(χ~2=6.741,P=0.009).According to multivariate analysis of Cox proportional risk regression model,β-catenin expression level,preoperative CA199 level,primary tumor size,and degree of tumor differentiation were the influencing factors of prognosis and survival(OR=0.139,9.883,6.117,3.834 P=0.001,0.001,0.009,0.028).Conclusions:β-catenin is highly expressed in PDAC,and its expression level is positively correlated with tumor size and TNM stage,which is a prognostic factor in PDAC patients. |
