Abstract/Summary:
Objective: Targett to study the effect of liver and kidney deficiency syndrome on the fibrosis of ligamentum flavum in patients with lumbar spinal stenosis,and to compare the expression levels of serum sex hormone,TGF-β1 and mi R-21 in patients with liver and kidney deficiency syndrome of lumbar spinal stenosis and non liver and kidney deficiency syndrome of lumbar spinal stenosis Objective to investigate the relationship between the expression of mi R-21 and TGF-β1 in lumbar spinal cord tissues and deficiency of liver and kidney,and to provide experimental research basis and reference ideas for TCM diagnosis of lumbar spinal stenosis and future TCM intervention.Methods: 1.According to the modern medical diagnostic criteria of lumbar spinal stenosis,TCM syndrome differentiation criteria of liver and kidney deficiency syndrome,inclusion and exclusion criteria of control research,51 patients with liver and kidney deficiency syndrome of lumbar spinal stenosis and non liver and kidney deficiency syndrome of lumbar spinal stenosis were selected for surgical treatment.The general information of the two groups was collected by electronic medical record system Venous blood was extracted from the two groups,and the sex hormones of the two groups were detected by enzyme-linked immunosorbent assay;a small amount of ligamentum flavum tissue was collected during the operation,and the expression level of TGF-β1protein in the ligamentum flavum tissue of the two groups was detected by Western blot,and the mRNA and miRNA-21 of TGF-β1 in the ligamentum flavum tissue of the two groups were detected by real-time fluorescent quantitative polynucleotide chain reaction According to the results of the experimental data,the expression levels of six sex hormone indexes,TGF-β1 protein,TGF-β1 gene and mi R-21 in the ligamentum flavum tissue of the two groups were evaluated;3.SPSS20.0 was used to analyze the experimental data,,and t-test was used if two independent samples were subject to normal distribution and homogeneous variance,and the measurement data were in the "mean ± standard deviation" table If the distribution is not normal,the rank sum test is used,and the measurement data is expressed as "median ± quartile interval".The counting data were compared by chi square test.P < 0.05 was statistically significant,P < 0.01 was statistically significant.Results: 1.The age of liver and kidney deficiency syndrome group(69.03±9.18)was higher than that of non liver and kidney deficiency syndrome group(50.00±11.96),P<0.01,with significant statistical difference;the course of disease of liver and kidney deficiency syndrome group(22.29±0.05)was longer than that of non liver and kidney deficiency syndrome group(11.32±0.07),P< 0.01,with significant statistical difference.2.It was found that the expression of luteinizing hormone and pituitary prolactin in male and female patients was higher than 0.05 in liver and kidney deficiency syndrome group and non liver and kidney deficiency syndrome group,and the difference was not statistically significant;the expression of testosterone in female patients in liver and kidney deficiency syndrome group(22.43±1.64)was lower than that in non liver and kidney deficiency syndrome group(26.59±0.92),P<0.01;the expression of testosterone in male patients with liver and kidney deficiency syndrome group(22.43±1.64)was lower than that in non liver and kidney deficiency syndrome group(26.59±0.92),P<0.01;The expression of testosterone in liver and kidney deficiency syndrome group(298.57±15.87)was lower than that in non liver and kidney deficiency syndrome group(532.47±22.72),P< 0.01;the expression of progesterone in female patients(0.32±0.05)was lower than that in non liver and kidney deficiency syndrome group(0.64±0.01),P<0.01;the expression of progesterone in male patients(0.61±0.05)was lower than that in non liver and kidney deficiency syndrome group(0.61±0.05)The expression of serum estradiol in female patients with liver and kidney deficiency syndrome group(12.42±2.86)was lower than that in non liver and kidney deficiency syndrome group(32.38±5.04),P<0.01;the expression of serum estradiol in male patients with liver and kidney deficiency syndrome group(15.84±2.92)was lower than that in non liver and kidney deficiency syndrome group(25.72±3.82),P<0.01;the follicle stimulating effect in female patients was better than that in non liver and kidney deficiency syndrome group(25.72±3.82),P<0.01;The expression of follicle stimulating hormone in liver and kidney deficiency syndrome group(29.84±1.34)was higher than that in non liver and kidney deficiency syndrome group(12.26±0.88),P<0.01;the expression of follicle stimulating hormone in male patients with liver and kidney deficiency syndrome group(12.73±0.48)was higher than that in non liver and kidney deficiency syndrome group(7.12±0.16),P<0.01.3.The expression level of TGF-β1 protein in liver and kidney deficiency syndrome group(0.86±0.07)was higher than that in non liver and kidney deficiency syndrome group(0.69±0.05),P<0.05;the expression level of TGF-β1 mRNA in liver and kidney deficiency syndrome group(3.78±0.26)was higher than that in non liver and kidney deficiency syndrome group(2.87±0.05),P<0.05.4.The expression level of miRNA-21 in liver and kidney deficiency syndrome group(0.088±0.010)was higher than that in non liver and kidney deficiency syndrome group(0.028±0.008),P<0.01.Conclusion: 1.The age of the patients with liver and kidney deficiency syndrome of lumbar spinal stenosis was significantly higher than that of the patients without liver and kidney deficiency syndrome.The course of disease of the patients with liver and kidney deficiency syndrome was significantly longer than that of the patients without liver and kidney deficiency syndrome;2.It is speculated that the level of sex hormone secretion can indirectly reflect the state and degree of liver and kidney deficiency;3.Liver kidney deficiency syndrome and TGF-β in lumbar spinal stenosis-β The expression level of miRNA-21 was positively correlated with the liver and kidney deficiency syndrome of lumbar spinal stenosis.Therefore,it is speculated that the liver and kidney deficiency syndrome of lumbar spinal stenosis is positively correlated with the proliferation and hypertrophy of ligamentum flavum to a certain extent;4.Based on the experimental results of this experiment,it is speculated that the liver and kidney deficiency syndrome of lumbar spinal stenosis can affect or even accelerate the changes of ligamentum flavum fibrosis.
