| Objective:To study the effects of tonic Chinese medicine and bao-yuan decoction on the life span of c.elegans and its related indexes,and to explore its mechanism.Methods:Single-flavored Chinese medicine and compound were prepared by extraction of two methods:alcohol extraction and water extraction.Taking Caenorhabditis elegans as an animal model,a crude drug extract group(50,100,200,500 μg/ml,1 mg/ml),a solvent control group and a blank control group(0 mg/ml)were established to observe the replenishing alcohol/water.The effects of extracts and Baoyuan Decoction on the lifespan and physiological indexes(reproductive ability,exercise capacity,body bending times,and head swing frequency)of N2 wild-type C.elegans.Through the oxidative stress test and heat stress experiment of nematodes,the mechanism of anti-aging drugs and Baoyuan decoction to delay aging was discussed.Results:(1)Life span test:The average lifespan of nematodes in the ethanol extract of Panax ginseng was significantly prolonged by 15.4%(200μg/ml),14.88%(500μg/ml)and 19.31%(lmg/ml),respectively(P<0.05).The average lifespan of nematodes in the extract of Astragalus membranaceus was significantly prolonged by 11.78%(200μg/ml group),18.78%(500μg/ml)and 19.26%(lmg/ml),respectively(P<0.05);The average lifespan of nematodes was significantly prolonged by 18.91%(100μg/ml group),20.01%(200μg/ml group)and 21.72%(500μg/ml)(P<0.05).The average lifespan of C.chinensis extracts was significantly prolonged.They were extended by 13.71%(200μg/ml),22.89%(500μg/ml)and 17.91%(lmg/ml)respectively(P<0.05).The average lifespan of nematodes in the extract of Angelica sinensis was significantly prolonged,which was extended by 13.91%(200μg)./ml),14.45%(500μg/ml)and 20.23%(lmg/ml)(P<0.05);the average lifespan of nematodes in the extract of Ophiopogon japonicus was significantly prolonged by 12.06%(200μg/ml)and 15.59(500μg/ml)and 14.81%(1mg/ml group);the average lifespan of nematodes in Baojitang alcohol extract group was significantly prolonged,respectively,extended by 20.99 100μg/ml),18.03%(200μg/ml)and 14.71%(500μg/ml)(P<0.05);the average lifespan of nematodes in the aqueous extract of Panax ginseng was significantly prolonged,extending 8.19%(200μg/ml)and 13.03%respectively.(500μg/ml)and 15.07%(1mg/ml)(P<0.05);the average lifespan of nematodes in the aqueous extract of Astragalus membranaceus was significantly increased by 8.94%(200μg/ml group),15.93%(500μg/ml group)and 17.61%.(1mg/ml group)(P<0.05);the average lifespan of nematodes in the licorice aqueous extract group was significantly prolonged,and the average survival time was extended by 19.58%(200μg/ml group)and 14.35(500μg/ml)respectively(P<0.05).The average lifespan of C.elegans was significantly prolonged by 19.74%(200μg/ml),19.11%(500μg/ml)and 14.01%(1mg/ml)respectively.The average lifespan of nematodes in the water extract of Angelica sinensis was significantly prolonged.It was extended by 12.63%(200μg/ml),15.77%(500μg/ml)and 20.91%(1mg/ml)respectively(P<0.05).The average lifespan of nematodes in wheat winter water extract group was significantly prolonged,and the average survival time was prolonged.17.05%(200μg/ml),17.32%(500μg/ml)and 14.84%(1mg/ml)(P<0.05);Baoyuan Decoction The average lifespan of nematodes was significantly prolonged by 17.08%(100μg/ml),20.65%(200μg/ml)and 18.23%(500μg/ml),respectively.The average lifespan of nematodes in the solvent control group was different from that in the blank group.Statistical significance(P>0.05).(2)Reproductive ability experiment:There was no significant difference in the number of eggs laid by nematodes in each drug-administered group compared with the blank group.(P>0.05).(3)Exercise ability test:After 6 days of drug treatment,ginseng,astragalus,licorice,cinnamon,angelica,maidong,Baoyuan decoction/water crude extract in a certain dose range(200,500μg/ml,1mg/ml)Both can significantly improve the ability of nematode transport,and the number of crawling peaks of nematodes increased significantly in 1 min(P<0.05).There was no significant difference between the solvent control group and the blank group(P>0.05).(4)Head swing frequency test:After 6 days of drug treatment,ginseng,astragalus,licorice,cinnamon,angelica,yoghurt,Baoyuan decoction/water crude extract in a certain dose range(200,500μg/ml,1mg/ml),the frequency of head worm swing was significantly increased,the number of swings was significantly greater than that of the control group(P<0.05),and there was no significant difference between the solvent control group and the blank group(P>0.05).(5)Body bending frequency test:There was no significant difference in the body bending frequency between the experimental groups and the blank control group(all P>0.05).(6)Heat stress test:Compared with the blank control group,the lifespan of the drug group was significantly prolonged.When the drug dose was 200,500 μg/ml and 1 mg/ml,the survival rate of the first,second and third antennas in the drug-administered group was significant.Compared with the control group(P<0.05),there was no significant difference in the average lifespan of the nematode in the solvent control group compared with the blank group(P>0.05).(7)Oxidative stress test:Compared with the blank control group,the lifespan of the drug group was significantly prolonged.When the drug dose was 200,500 μg/ml,1 mg/ml,the drug group survived in the first,second,third,and fourth antennas.The rate was significantly higher than that of the control group(P<0.05).There was no significant difference in the average lifespan of the nematodes in the solvent control group compared with the blank group(P>0.05).Conclusion:Alcohol/water crude extracts such as ginseng,astragalus,licorice,cinnamon,angelica and Ophiopogon japonicus can effectively prolong the average lifespan of nematodes;Baoyuan decoction(ginseng,astragalus,licorice,cinnamon)alcohol/water thick The extract can effectively prolong the average lifespan of the nematodes and has a better anti-aging effect.Its mechanism of delaying aging may be related to the ability of drugs to improve the anti-stress ability of nematodes. |
