Objective:To predict the underlying mechanism in treating MCI model mice of the sniff-form BingChangSan by means of network pharmacology,behavior and pathology.Methods:(1)With extracting the volatile oil and detecting by GC-MS,filtering out the main odorants of BingChangSan,predicting their targets and the signaling pathways in treating MCI by ordorant with OR;(2)The sniff-form BingChangSan was prepared by adding borneol,and after two or four weeks of intranasal administration,its effects were detected that on the nesting behavior and Morris memory of MCI model mice,and the changs of TNF-α in serum,together with A(3 and caspase 3 in the hippocampus of MCI model mice,etc;(3)Detecting the protein expression of SIRT1/NF-κB in the hippocampal of MCI model mice by western blotting.Results:(1)Seven odorants were obtained,all with high BBB permeability;The enrichment analysis identified two pathways may be most relevant to the mechanism in treating MCI by sniffing BingChangSan based on OR,that is,cAMP signaling pathway and PI3K-Akt signaling pathway;(2)BingChangSan can improve the ability of MCI model mice on nesting behavior and recalling Morris long-term memory,and reduce the production of TNF-α in serum,together with Aβ and caspase 3 in the hippocampus of MCI model mice;(3)BingChangSan can up-regulate SIRT1 and down-regulate NF-κB expression in the hippocampus of MCI model mice;Conclusion:The sniff-form BCS can effectively improve the daily life and spatial memory ability of MCI mice,the mechanism may be associated with inhibiting apoptotic factors such as TNF-α、Aβ、caspase-3 by SIRT1/NF-κB signaling pathway.
