| Objective:To study the specific mechanism of Peiben Gushu Recipe(PBGSF)by promoting bone formation to intervene in PMOP,combined with the results of previous studies,to improve the mechanism of PGBSF in preventing and treating PMOP.Methods:Conclusion:Conclusion PGBSF can promote the proliferation and maturation of osteoblasts and inhibit the apoptosis of osteoblasts to play its role in promoting osteogenesis,and its effect is dose-dependent within a certain range(P<0.05).The mechanism of action may be related to the promotion of Akt/PPAR-y Signal pathway activity is related.Method:36 SPF-grade 6-month-old non-pregnant female healthy SD rats were randomly divided into 6 groups,namely:sham operation group,model group,low-dose group of Peibengushu prescription,medium-dose group of Peibengushu prescription,and training group.Bengushufang high-dose group and western medicine group.After 1w adaptive feeding in separate cages under natural light,a model was created,and a bilateral egg removal method was used to establish a postmenopausal osteoporosis rat model(the rats in the sham-operated group were removed 1g of fat beside the ovaries).In 7 days,80,000 U/d of penicillin was injected intramuscularly through the hind limbs to prevent infection.The equivalent dose of rats was calculated as 6.3 times for adults.The administration was started 4 weeks after the model was built,and the administration volume was 0.5ml/100g once a day for 8 weeks.Peibengushu prescription low,medium and high dose groups:Prepeibengushu prescription 5,10,20g crude drug/(kg·d);Western medicine group:alendronate sodium 0.1mg/100g;model group,Sham operation group:saline 0.5ml/100g was administered.After the last administration,fasting for 24 hours to obtain materials,determine bone mineral density(BMD),bone tissue elastic displacement,limit displacement,bone tissue fragility,bone cell mineralization level,bone tissue Akt/PPAR-γ protein expression level,serum ALP content,Changes of oxidative stress index ROS level.Result:Compared with the sham operation group,the BMD,bone elasticity and ultimate displacement,Akt expression level of the model group decreased(P<0.01),fragility increased(P<0.05),no calcified nodules appeared in the bone tissue,and the expression of PPAR-y protein The level,serum ALP and ROS all increased significantly(P<0.001).Compared with the model group,the BMD of each treatment group increased(P>0.01);the elastic displacement of the PBGSF low and medium dose groups increased,and the fragility was significantly reduced(P<0.01);the high dose group and the western medicine group increased the elastic displacement and ultimate displacement significantly.Significantly(P<0.001),and the brittleness decreased(P<0.05).A large number of calcified nodules were seen in the Chinese and Western medicine groups,and the calcium deposition in the bone tissue of the Chinese medicine group was positively correlated with the administration concentration.The Akt protein expression levels of PBGSF low and medium dose groups increased(P<0.05),and the Akt protein expression levels of the Peibengushufang high-dose group and alendronate sodium group were significantly increased(P<0.01).The expression level of PPAR-y protein in bone tissue of the low-dose group of Peiben Gushu Decoction decreased significantly(P<0.01),and the expression level of PPAR-y protein in the middle,high-dose group and the alendronate sodium group of Peiben Gushu Decoction decreased significantly(P<0.01).0.001).ROS in each treatment group was significantly reduced(P<0.001),and the effect of the traditional Chinese medicine group was better and concentration-dependent.Conclusion:1.Peibengushu Decoction can effectively improve postmenopausal osteoporosis,and its effect is dose-dependent within a certain range.2.Peibengushu Decoction can promote the proliferation and maturation of osteoblasts to play its role in promoting osteogenesis.3.Peiben Gushu Decoction can improve osteoporosis through Akt/PPAR-y pathway and anti-oxidative stress. |
