| Based on the unique symbiotic relationship between Gastrodia elata and Armillaria sp.,the co-culture of the associated fungi of G.elata and Armillaria sp.would discover secondary metabolites with novel structures and biological activities.The secondary metabolites of co-culture of two fungi(Perenniporia sp.DS-8 and Penicillium sp.DQT-68)isolated from rhizosphere soil of Gastrodia elata with Armillaria mellea were systematically studied and their activities were also evaluated respectively.The thesis was divided into three chapters.In chapter one,the research progress of chromone compounds and their biological activities in the secondary metabolites of microorganisms were reviewed.In chapter two,co-culture of the fungus Perenniporia sp.DS-8 from the rhizosphere soil of G.elata with Armillaria sp.led to the production of 9 secondary metabolites,including perenranone(1),diwortmannine(2),perenranter(3),perendenone A(4),perendenone B(5),4-hydroxy-isobenzofuran-1(3H)-one(6),7,8-Dimethylalloxazine(7),Diorcinol(8)and Wortmannine G(9).Then,the cytotoxic activities and antiinflammatory activities of the new compounds(1-5)were investigated.In chapter three,co-culture of the fungus Penicillium sp.DQT-68 from the rhizosphere soil of G.elata with Armillaria sp.resulted in the production of 11 secondary metabolites,including a new compound vascouran B(1),together with ten known ones vascouran A(2),canescone F(3),O-menthylcurvulinic acid(4),methyl 2-acetl-3,5-dihydroxyphenylacetate(5),ethyl 2-acetyl-3,5-dihydroxyphenylacetate(6),streptokordin(7),integrastatin B(8),(22E)-5a,8a-epidioxyergosta-6,22-dien-3b-ol(9),5-O-methylmelledonal(10)and Melledonal A(11).Then,the AChE(acetylcholine lipase)inhibition activities,cytotoxic activities,anti-inflammatory activities and PTP1B(protein tyrosine phosphatase 1B)inhibition activities of compound 2 was investigated.As the results,compound 2 showed moderate inhibition activities to AChE. |
