| Prostate cancer(PCa)has become a"killer"threatening the health of elderly men worldwide.The occurrence and development of early-age prostate cancer depends on the stimulation of androgen.Androgen can stimulate and regulate the abnormal expression of genes and proteins in prostate cancer cells.Studies have shown that androgen can also affect the abnormal expression of some long non-coding RNA(lnc RNA),which is related to the occurrence and development of PCa.Previous results from our own research showed that a large number of lnc RNAs were abnormally expressed in RNA-seq data,with the treatment of five different concentrations of androgen to the AR+(AR,Androgen receptor)prostate cancer cell line LNCa P.We also identified a novel clinically relevant,DHT-regulated lnc RNA in the prostate cancer cells via analysis of our data against the TCGA database.The expression of it is ddifferent in normal prostate and malignant prostate cancer,and patients with high expression had a high-grade PCa according to the pathological grade(Gleason grade)of clinical data,suggesting that the lnc RNA functions in occurrence and development of prostate cancer.Because this lnc RNA is located in the fourth intron of Centromere protein N(CENPN),we named it lnc CENPN-IT(IT region of the Centromere Protein N),according to the naming rules.Next,we analyzed the effect of lnc CENPN-IT on the proliferation,migration,apoptosis and cycle of prostate cancer cells.First,we used real-time fluorescent quantitative PCR technology to verify the expression of lnc CENPN-IT in AR+prostate cancer cells and verified that lnc CENPN-IT was up-regulated by DHT,and that the most significant change was found in LNCa P cells.Then we performed our in vitro experiments with LNCa P cells.Indeed,in vitro loss-of-function studies revealed that knock-down of lnc CENPN-IT inhibited proliferation and migration,promoted cell apoptosis,delay cell cycle,whereas overexpression of lnc CENPN-IT promoted the proliferation and migration,accelerate the cell cycle processin in LNCa P cells.In order to further explore the role of lnc CENPN-IT in prostate cancer cells,we firstly examined the distribution of lnc CENPN-IT by using fluorescence in situ hybridization(FISH).The results showed that lnc RNA mainly located in the cytoplasm of LNCa P cells.Secondly,we performed GSEA,GO and KEGG enrichment analysis on the information against the Cancer Genome Atlas(TCGA)database,and found that lnc CENPN-IT was mainly enriched in centromere protein,Cycle,cancer-related pathways.In summary,we have identified that a previously unannotated lnc RNA:lnc CENPN-IT was upregulated by androgen DHT in a dose-dependent manner.Clinical data showed that it was highly expressed in prostate cancer tissues and was closely related to the progression of prostate cancer.lnc CENPN-IT may regulate the proliferation,migration,apoptosis and other cell functions of prostate cancer by affecting pathways such as cancer,centromere proteins,and cycle apoptosis,thereby having an influence on to the occurrence and development of prostate cancer.This study extends our understanding of the pathogenesis of prostate cancer and provides a potential target for the treatment of prostate cancer. |
