Objective: 1.To detect the expression of circPGM5 in bladder cancer tissues and cells;2.To explore the effect of overexpression of circPGM5 on the proliferation of bladder cancer cells;3.To explore the effect of overexpression of circPGM5 on the migration function of bladder cancer cells.Methods: 1.Whether circPGM5 is circ RNA was proofed by agarose gel electrophoresis;2.the expression of circPGM5 in bladder cancer tissues and cell was finded by real-time quantitative PCR(QPCR)technique;3.Bladder cancer cells were solidly transfected with lentivirus,and the efficiency of infection was detected by q PCR;4.The effect of overexpression of circpgm5 on the cell cycle of bladder cancer cells(T24 and EJ)was detected by flow cytometry;5.The effect of overexpression of circpgm5 on the proliferation of bladder cancer cells(T24 and EJ)was detected by cloning of cells,CCK8 and edu test;6.The effect of overexpression of circpgm5 on the migration of bladder cancer cells(T24 and EJ)was detected by cell scratch test.Results: 1.Different primers were designed to amplify circPGM5,and the resulting products were tested by agarose gel electrophoresis.Sanger sequencing was performed on the q PCR products of circPGM5 to verify the formation of head-to-tail splicing covalent binding;2.According to q PCR,circPGM5 was lower expressed in bladder cancer cells(T24,EJ,and BIU87)and bladder cancer tissues compared to normal urothelial cells(SV-HUC-1)and paracancerous tissues;3.According to q PCR,circPGM5 was significantly upregulated by lentivirus overexpression in T24 cells and EJ cells;4.Flow cytometry results show that overexpression of circPGM5 can block bladder cancer cells(T24 and EJ)in G1 phase;5.Plate cloning,CCK8 and edu experiments show that increasing the expression of circPGM5 can significantly prevent the proliferation of bladder cancer cells(T24 and EJ);6.The results of cell scratch study showed that the function of bladder cancer cells(T24 and EJ)was not affected by increasing the expression of circPGM5.Conclusion: 1.The expression of circPGM5 is low in bladder cancer tissues and cells;2.Increasing the expression of circPGM5 can significantly prevent the proliferation of bladder cancer cells;3.Overexpression of circPGM5 does not affect the migration of bladder cancer cells;4.The role of circPGM5 in bladder cancer provides potential biomarkers and therapeutic targets for the treatment of bladder cancer.
