Objective: Summarize the research status of Epimedium Epimedii,The chemical components of Epimedium and Epimedium Epimedii are extracted and separated.The chemical components of 12 batches of Epimedium Epimedii were compared.To study the protective effects of Epimedium and Epimedium Epimedii on Kidney Injury Rats.Methods: 1.Optimize the different extraction solvents of flavonoids of Epimedium and Sunflower Epimedium,It is established that 70% ethano-waterl is used as a solvent and heated to reflux to obtain crude extracts of flavonoids of Epimedium and Epimedium.The crude extract was depressurized,distilled,chloroform extraction degreasing,polyamide resin column adsorption,70%ethanol-water,and the total flavonoid content and yield of Epimedium and Sunflower Epimedium were calculated.The crude extracts of epimedium and roasted epimedium are used as a spare for the following pharmacological activity experiments;Extraction of polysaccharides from Epimedium and Epimedium by water extraction and alcohol precipitation,The Seavge method removes protein,and the total polysaccharide after removing protein is washed with various solvents,Refined epimedium and sunburned epimedium polysaccharide ingredients,Calculate the content and yield of total polysaccharides of epimedium and sunburned epimedium.2.The extraction solvent,extraction method,extraction time and extraction times of the flavonoids of Epimedium Epimedii are optimized;3.HPLC method uses acetonitrile(A)-water(B)as mobile phase to detect the content of 7 flavonoids in Epimedium Epimedi and conduct methodological investigation,and establish the fingerprint of the flavonoids of Epimedium and Epimedium Epimedi4.The flavonoids of Epimedium Epimedii were analyzed by one measurement and multiple evaluation.Icariin was used as the reference peak to calculate the relative correction factors of the other6 components.The detection results of the external standard method and the one measurement and multiple evaluation method were compared.5.The contents of 7 flavonoids in Epimedium Epimedii were set as variables for multi-stoichiometric analysis;6.Establish a rat kidney injury model,conduct behavioral observations on rats,measure body weight,and detect the effects of Epimedium and Epimedium Flavonoids on serum levels,renal function indicators,and blood routine factors in rats with renal injury,and compare the changes of pathological sections to evaluate the protective effect of Epimedium and Epimedium total flavonoids on kidney injury in rats.Results:1.Quantitative analysis of total flavonoids and total polysaccharides in epimedium,Epimedium Epimedi scorched and isolated Epimedium sylvestris and epimedium scorched,showed that the average content of total flavonoids was 6.87% and6.75%,respectively.The average yields were 12.66% and 12.15%;the average content of total polysaccharide components were 4.89 mg/g,4.35 mg/g,and the yields were 3.65%and 2.48%.Compared with roasted Epimedium Epimedi,the content and yield of flavonoids and polysaccharides in epimedium are higher.2.Weigh 250 mg of sample powder that has passed through a 24-mesh sieve,transfer all to the Erlenmeyer flask,Add 70% ethanol-water solution,25 m L,ultrasonic extraction for 30 min,Bring the extracted solution to room temperature,weigh it,use 70% ethanol-water to make up for the weight lost during the extraction process,and mix the solution to get the required test solution.An HPLC method for the detection of flavonoids in Epimedium Epimedi was established,and the 7 chromatographic peaks were identified as Epimedin A,Epimedin B,Epimedin C,Icariin,Baohuoside I,and Baohuo.Glycoside II,Icariin I,and established a fingerprint of the flavonoids of raw and roasted epimedium.The similarity of 12 batches of epimedium and 12 batches of Epimedium Epimedi was greater than 0.92.The results of one test and multiple evaluations suggest that the relative correction factors of the other 6 components calculated with subicariin as the reference substance are1.389,1.401,1.494,0.839,1.524,0.881 and the reproducibility of the correction factors is investigated,RSD The values are all less than 2%,indicating good reproducibility,and there is no significant difference between the external standard method and the QAMS calculated value.3.After modeling,the weight decreased significantly,the extension was dull and dull,and the modeling was successful.The weight of the administration group decreased slightly and the eyes were brighter than that of the cisplatin alone administration group;after the modeling,the rats UA,CRE,BUN,MDA,NO,NEUT% The content of LYMPH%increased significantly,which was significantly different from that of the blank group(P<0.05),and the content of SOD and MONO% was significantly reduced,which was significantly different from that of the blank group(P<0.05);all indicators of drug intervention were improved,The high-dose group of Epimedium Epimedi had better effect.Conclusion:After extraction and separation of flavonoids and polysaccharides in Epimedium and Epimedium Epimedi,the content and yield of flavonoids and polysaccharides in Epimedium are higher than that of Epimedium Epimedi;Fingerprinting combined with one test and multiple evaluations qualitatively and quantitatively analyze the epimedium,and comprehensively evaluate the epimedium;After cisplatin was used to successfully create membranes in rats with kidney injury,each dose of Epimedium and Epimedium Epimedi had protective effects,and the high dose of Epimedium Epimedi had the best protective effect. |
